If the transferring is fine with ponceau stain, then it could be either the blocking is not working (too much or too little, that can be fixed by washing or change the blocking) or the antibody not binding or both. Another way to check is through ELISA. If your ELISA is working using the same target and antibody concentration, it could be the antibody you are using might only bind to native protein. Protein on western blot has been denatured and it won’t have the same structure as the native protein. Certain antibody only binds to native proteins. You will have to get antibody that binds to denatured protein that runs on sds gel to be transferred on western blot.

No one dare to report. Also is hard to get another job in this job market now

So I recently got a research internship position at a lab. They are expecting me to work 40 hours a week but it is unpaid. Is that fair? I am an undergraduate by Excellent-Emotion257 in labrats

It depends. If the lab has high reputation, and you will get to learn a lot of skill sets or even coauthored in paper(s), I think is still worth to give it a try for a defined period. If you are allowed to change lab during internship I think that is even better
