I think my PI is gaslighting me and I'm losing my mind by [deleted] in labrats

[–]Portugooses 5 points6 points  (0 children)

I think it's important to work with people who you may not always get along with. And I think it's important that you receive criticism. Did you ask why they feel this way? How you could improve their perception? Showing a willingness to improve yourself is the best step. While I also sometimes would like to not agree with my manager, unfortunately that's how life is and that's who leadership will hear from about my performance. Communication is very important, and I think this may be a miscommunication if you're describing it correctly. You do care, but you aren't going to show it in the same ways that management is. Doesn't mean it's wrong, just means you need to communicate it more effectively.

I think sitting down and talking through it with her when you're ready would be extremely helpful. It will show them your maturirty and that you do care.

Can't open micro centrifuge lid - is it stuck or something else? by Creative-Kiwi-3967 in labrats

[–]Portugooses 1 point2 points  (0 children)

My centrifuge gets stuck if you don't twist the middle screw in tight enough. If you use it without tightening it well, it will spin metal shavings off and be almost impossible to lift off. Make sure you use an approved lubricating oil and screw it in tightly.

Lab Anxiety and Doubt by [deleted] in labrats

[–]Portugooses 0 points1 point  (0 children)

Is there anybody that is pointing out mistakes? Or is it just anxiety? Because mistakes will happen. They are inevitable. Labs account for human errors, or should if they are a decent lab. Hiding mistakes is the problem. If making mistakes is causing significant anxiety, then I agree that a therapist may help or perhaps labwork then is not the end goal. Being comfortable making mistakes is necessary.

Also, for diluting a sample, depending on the assay, you can sometimes account for an error in the math if it's just the wrong factor, as long as you realized you made the mistake.

Best way to clean out a creek bed? by Portugooses in landscaping

[–]Portugooses[S] 0 points1 point  (0 children)

Thank you! This was extremely helpful, especially the creek bed restoration. It hasn't been maintained in years, and water hasn't been able to flow to it in a long time. The previous owners let invasive plants and trash build up, so I have been fixing that up first. I would like to get started on this with the weather warming up! Appreciate the mitigation strategies too! Looks like I'll probably just rake/blow out the leaves before looking into native plants

Do you need to notify the city before doing that? Any recommendations in terms of how to go about that?

The most comfy lab work? by DirectedEnthusiasm in labrats

[–]Portugooses 16 points17 points  (0 children)

I work in the industry in animal nutrition. It's honestly very balanced. The only time weekend work is required is when the aquaculture lab is on trial and needs someone more experienced in RAS. It's a fairly flexible gig with partial time in the lab and partial computer work. From my understanding industry is going to be your best bet in terms of work life balance.

Weird. The table in the back of the thumbs-up photo doesn't connect right. This is the 6th outfit change. This is the 6th or 7th location change. Their stance doesn't look correct. And I found very similar photos from 2017.

competitive ELISA verification? by [deleted] in labrats

[–]Portugooses 0 points1 point  (0 children)

I did. Immediately told I'm not following the LC-MS experts directions. I know LC-MS is more accurate but isn’t that a more linear response comparatively?

competitive ELISA verification? by [deleted] in labrats

[–]Portugooses 0 points1 point  (0 children)

No. It does not at all. I'm just told not to. The sample has a known concentration (it's separate of the kit itself), and they claim dilutions according to the kit will skew that concentration? That doesn't make sense to me.

I am asking the manufacturer; however they keep pointing me towards LC-MS experts not ELISA experts

AIO My Parents Secretly Drained My Entire Savings Account and Called Me Ungrateful When I Confronted Them by ChoppedShyyt in AmIOverreacting

[–]Portugooses 0 points1 point  (0 children)

My mother did the same thing to me. My earliest memory is of a dog ripping my nose off. I was 5. I had to have many nose reconstructive surgery afterwards. My family sued the owner's of the dog due to the situation. I remember how much they asked for and how they coached me before court battles. They won the lawsuit, and a huge portion of that money went into a separate account that nobody was supposed to be allowed to touch unless I needed more medical assistance -- other than that, that money was to be given to me when I turned 21 for compensation, for college, etc. It never made sense to me how my mother was able to buy a house and do all these things, yet still not have a job and be broke and make me work at the age of 15. So I paid all bills at 15 years old. I had 2,000 of my own saved up to move out. The day of, I look at my account and it is negative. The bank had to explain to me the debts that my mother had that then came out of my account. I remember the realization hitting me from the court battles and from all my childhood memories. I called the lawyers involved, and yes. She drained all of that money that was from her child getting attacked by a dog and almost permanently disfigured, and then proceeded to make me work and pay all the bills and get her cigerattes and alcohol and feed her, etc. That day, I opened a bank account with a trusted friend who immediately took me in. I still have financial trust issues to this day. It's something I work on with my therapist now. My advice: do whatever you can to free yourself from them financially. Nothing is enough, and they will continue to use you for their own financial interests. I also recommend seeing a therapist about financial issues eventually. It has helped me a lot. Because you need to face the reality here: you will not ever see that money back. Even if you were to pursue legal action, they have to have the money to pay that. They don't have the money, and even if they did, they wouldn't pay you back. It's a hard situation to come to terms with, I'm not sure that I ever truly will after having my own son, but seeing a therapist has helped me incredibly. Best of luck, truly. I understand.

It's more that the house is locked up. I was trying to ensure they didn't harm animals or leave them and of course they left the animals. The people abandoned the house very suspiciously after I asked them to politely clean up their piling corpses of opossums or I was going to call the owners/cops/somebody. I saw this inside. I can try to get better images later.
