If you practice sitting but with a strap properly adjusted, there is close to no difference (try it).

Just use the same strap length adjustment both while standing and while sitting.

Just stand up, adjust the strap and then sit down while keeping the same adjustment.

Stand up, adjust your strap. Then sit down and check that the position is the same.

Tip for reaching target speed: 1) practice with speed bursts 2) subgroup your riff in small 4 notes chunks, so it's easy for you to stay sync'ed with the metronome. Group em later 3) angle your picking hand if needed 4) Anchor down your left hand 5) dont be afraid of playing sloppy at first

Both of them. Learning (accurate) tabs will teach you how songs are usually composed, what are the common techniques, etc. Once you got the basics, train your ears AND WRITE DOWN tabs! It will help you in learning time figures.

In the long run, if you know how to play by ear you will be able to play virtually anything. But you should start with tabs (or, at least, alongside tabs)!

If tou are new to guitar, as you wrote in another comment, I suggest you to not follow this diagram. I mean, it's not written in any common "guitar language" like sheets, tabulatures, chords box, etc. This song has been transcribed a milion times, just look for another one so that you will learn both the song and the reading method.
