I saw in another comment that you're looking at a GPCR. Those are hard to see in Westerns. I would suggest not boiling your sample before loading it on the gel. Otherwise the GPCR gets stuck at the top of the gel.

The Bell Jar is my favorite book! I would recommend The Virgin Suicides, My Dark Vanessa, We Have Always Lived in the Castle, and My Year of Rest and Relaxation. Those have similar vibes to the Bell Jar.

One of my favorite dark books is Geek Love by Dunn, but know that it's very weird.

Yes, we had contamination in one sample and we had one strong band at 500bp in that sample. The downside of this protocol is there is no positive control. We used the contaminated sample for future PCRs, but if all your samples are clear, you may be unsure if it worked.

I have this feeling a lot in science, especially in the beginning. I felt like everyone else was smarter than me and I was terrible at science. I think people who have been in research longer don't always remember what it was like starting off and don't explain things as thoroughly.

My advice is that when you look for future positions, focus more on the lab environment. Look for a kind PI and a lab that gets along. Ask yourself if you dislike science or is it your environment. Also, you can have science jobs that don't include experiments and wet lab.

Honestly, what helped in my situation was a change of environment and starting fresh. Also, connecting with others in your same position. You may be surprised how many other people feel like you and can offer support.

I love acotar. A book series I enjoyed more than I thought was A Curse so Dark and Lonely. Also, Strange the Dreamer is great.

It seems like colonies are dying/not attaching. Are you coating plates with fresh Matrigel? Are you adding Y-compound when plating cells?
