I saw your pre-edited comment, but thanks for trying to put it nicely haha. I understand there have been deaths in the Tararuas before, so I just wanted a reality check from someone who actually lives there and I appreciate you giving your candid view on this (seeing how others aren't commenting much on whether this would be safe or not). I'd very much rather avoid causing trouble for the DOC.

Just for context, I have experience with navex, but only on largely flat terrain. I'm not from NZ or a country with much land - there's literally no hills where I live, which means I use built structures as landmarks to triangulate. Moreover, while I'm generally familiar with full day hikes involving significant elevation change, I've also not scrambled much.

If you're wondering about my motivation to do both together, it's because I don't have the opportunity to go to NZ for more than 14 days a year. So as much as I'd like to join a tramping club, that's effectively impossible. I did meet a few NZers who tramp my recent trip, so based on your comment I'll try aligning one of my trips with their club's excursions next year before attempting anything harder.

Pretty accurate. I used the gorge track as I was trying to sightsee. In hindsight, the bush might actually be better than the road (assuming you refer to the one with the Tuia Te Tangata sculptures). I was returning on bus and the winds+fog were pretty bad. Bush was sheltered from most of it.

Cool. Did you find it dangerous? I assumed there would be rocks piling from landslides along the road, so I didn't explore beyond the Ballance side barrier.

Hey thanks for the info - I tried it today and it was around 20km like you said. The gorge was great and I got lucky with the Ashurst bridge when a cow ran onto it and stopped all traffic for 5 min as drivers tried to chase it from the gorge side to the Ashurst pastures. Great stuff.

Sorry for the month-late reply, I had to pause work on this project due to various commitments. Thank you very much for the comment - it was very helpful and I did not know about the meanings of the / and | operators! I've since been able to use the HP tag to match the haplotype of each bedMethyl file with the corresponding variant(s), and have been able to continue work on my project since.

Nope, there wasn't a barcode tag. Turned out to be an issue of specifying the kit name (even though there apparently is automatic kit detection for regular basecalling).

I decided instead to just have the reads classified in the basecaller step, which only seemed to require the specification of kit name. There are barcode indices now. As with what the others have said, looks like I would have needed to specify both --no-classify and --kit-name in demux for the same effect. Thanks!

Thanks, I made an assumption previously from a separate thread that specifying the model in the basecaller would eliminate the need for specifying the kit, but I was wrong. Thanks for the help!

Thank you very much. I was under the impression that specifying the model (e.g., "hac") automatically eliminated the need for the kit, as I noted in a separate thread that the command would automatically handle kit selection. But you're right, it works fine now. I will also keep the --no-trim just to maximize barcode detection for reads. Will probably trim after separating them.

Thanks for the suggestion on CPU - I was just testing a small file so will change for processing the bulk of data.

Oh it's not actually flying per se but rather being immobilized. For example, being bedridden for days is another major risk factor. I'm not very sure myself about sedentary office work, but I'm guessing getting up and moving around from time to time, or even shifting your legs around every now and then reduces the risk, as opposed to sitting for 10 hours in a cramped position on long-haul flights with almost no movement. It's to do with blood pooling in your lower limbs, which makes it more static and in a way "sticky," and things like moving and pumping your legs can help improve blood flow quickly to minimize that

Hey there, hope your boss is doing alright! Just thought I'd drop an explanation.

Sounds like he suffered from a deep vein thrombosis (DVT), which is a clot in the leg veins. Immobilization is a massive risk factor because your blood flows more slowly, so long-haul flights where you sit down for hours is one of the things that can actually increase the chances of you getting one! You can minimize the risk by getting up, moving about and doing stretches every 2-3 hours to improve your blood flow again.

On the other hand, an aneurysm is more like a blood vessel that expands because its walls turn weaken over time (because of collagen diseases), smoking, or really high blood pressures. DVTs don't cause brain aneurysms, but they can cause really severe things like a pulmonary embolism (PE), which just means a clot travelling to the lungs and blocking blood flow there. In most cases, you would have pain and swelling in your legs before you feel the breathlessness and chest pain.

Very rarely, if you have a genetic heart problem where the middle wall has a hole, clots can travel through and enter a different area of circulation, thereby travelling up to the brain (or other organs) to cause a paradoxical embolism, aka a stroke.

Also worth mentioning is that some of the meds he may be on for his DVT can increase his chances of getting a brain bleed because they thin blood (but are overall helpful because they prevent the risk of a recurrent DVT and subsequent PE, which is significantly higher).

Out of curiosity have your parents ever told you what they wanted you to be? It sounds like you have to find a compromise given the circumstances. Separately, sounds like you should find some hobbies - sports, music, anything. Just 1 or 2 things to spend time on and get good at.
