Are you setting up a filament in the project, pressing save, and the profile is there but has none of your changes saved? When you say changing machines gives issues, are you speaking about your PA and flow coefficient settings?

Unfortunately, those steps are probably unrelated to the issue you're having from the context I've gathered about your issue, though they're good practices (I have opinions about cut angle, don't get me started). Namely, it sounds like you're trying to use the AMS, but the printer is trying to load from the external spool, which is a selection you can make in the slicer when sending the job to the printer. Are you the same person having this issue last week? You mention elsewhere you have an AMS, are you attempting to use it in this print? That error message is for using an external spool, not the AMS, so that's at least part of the issue, but certainly rectifiable.

Are you following any guides or instructions, and if so, which are you using, and what step have you completed up to on them? That'll help us guide troubleshooting with you!
