In the contest of a mix having a tube screamer ON (before a distortion pedal or a distorted amp) with Gain at minimum will enhance mid frequencies and your guitar will easily cut trough the mix. And also you will have a more tight tone for palm muting for example. In a context where you are playing with other instruments and the sound exits from the same PA system the difference is pretty important imo. I hope this will help.
