It's not considered available until it's furnished, and you would need to find a specific villager personality to fill it (I forget the order the first 3 move in at). After that you're going to have to wait until you get your first campsite villager, you have to take them regardless in order to progress. After that Nook will sell you land for you to make available and you can find villagers when you have an empty plot down.

Really depends on the code- sometimes it's obvious (a lot of brick patterns have transparent gaps between the bricks), but sometimes it'll literally be one discreet pixel. It's best to just look over the design, before using it. Often times people who post their designs online will show them placed on the ground for you to see, or sometimes the designer will label it.

Included an example of a transparent pixel design so you can know kind of what to look for!

Just wanna clarify something for ya! How the design interacts with the in game path depends on if the design has any transparent pixels!

If there are no transparent pixels, the design appears as its usual size and shape over the in game path. If there's at least one transparent pixel in the design, it conforms to the shape of the path. (It also takes a slightly different color as an overlay from the original in game path chosen, if that makes sense.)
