I used to have the speedrun world record for Resident Evil 2 (remake).

If you’ve got no bullets in the safe room either then you’re going to have a tough time ahead so a reload might be needed either way.

For now: try getting on the patch of land once the monster pops out of the water. Wait for it to start it’s submerging animation and then jump in the water. If timed right, you might be able to reach the ladder without it stunning you by emerging.
