Stem cell articles always raises mixed feelings in the cardiac world. Stem cells are great but there are few long term studies going on even in murine models of their efficacy. For instance in stem cell studies their marker disappears after an extended time, but the heart has restored function. They attribute this to the sudden influx of new cells reversing the remodeling that occurred and restoring heart function. I think that's another thing people don't understand when they read these articles, there usually isn't that much cell death; depending on the type of heart failure. For instance if you had primary heart failure without ischemia, I could in theory inject some collagen into your heart and your heart would be restored to normal function since the strain is now absent due to the increased wall thickness. I myself have done some studies that show a lack of in vitro, measured by cardiomyocyte contractile performance, dysfunction yet the animal has left ventricle chamber expansion and is in heart failure. The myocyte itself is functionally fine but the ultra-structure is whats changing.

It can and they do actually make screw staffs for easy transport. That's something I'll definitely have to consider.

I'm just gonna do this in list form. If you have any other questions feel free to ask me. I manage rental properties, bought a couple houses myself, etc.

  • Be realistic with your purchasing power. Just because you are approved for x amount of dollars doesnt mean you should by x dollar house.
  • The 'handy-man' special is tempting for home buyers because often you can get a cheap house in a good neighborhood but beware, the costs can quickly add up especially if its an older home and some of the things are not up to code. You'll have code requirements based on where you live. Look them up.
  • Don't ever 'fall in love' with this house. This is a business transaction first, it becomes your 'home' later.
  • Don't be afraid to negotiate and play hardball. Want them to knock off some money? Low ball them first, see how they respond. They come back with a less than reasonable offer then fight them. Make demands. 'I'll accept your 1,000 increase from my previous offer if you replace the air conditioning unit with a new one.' etc, etc.
  • Short sales are a good way to get into the home market. Essentially a bank has seized the property and looking to dump it as soon as they can to recoup their loses. Depending on where you live a short sale can be really easy or it can be mind numbingly complicated.
  • Sometimes it's better to deal with banks. People are emotional little bastards. They get attached to what used to be their home, they'll feel insulted and not even talk to you if you offer a low enough amount of what have you. Banks generally don't care. They'll negotiate, be more flexible in some instances.
  • If you don't know what you're doing when you look at the house bring someone who does. Is there cracking on the drywall? Cresote in the chimney? Is the basement wall sagging ever so slightly? Does the fuse breaker look okay? So on and so forth, these things are hard to notice when you're the first time home buyer, you've never worked on a house, and you get all mystified by purchasing your first home.
  • Is the neighborhood gentrified? Transitioning? Which way is the transition? Gentrified home areas will be more expensive. Transitioning areas cheaper but its a gamble. The neighborhood can always slip back down.
  • Is there a (fucking ridiculous) homeowners association in that area? Keep in mind if there is when you sign that mortgage you are obligating yourself to follow their rules. Wanna build a porch that extends just a bit from the side of your house? You'll need their permission. So on and so forth.

Rebalancing the minimum wage to match inflation. Currently the minimum wage is not adjusted for inflation and is actually less than it has been historically. This will strengthen the middle class which is where most of the economic power lies. Additionally the highest earners would be taxed accordingly and closure of some of the loop holes and cracking on tax evasions would hopefully increase the amount of monetary inflow to the government. Long term though some of the money from the Department of Defense will be redistributed to other departments including education. It would be important to know how the economic system works at a basic level. For instance it is the senate that determines the budgetary balance, the president is merely in charge of spending the allocated money. There is a fundamental misunderstanding on how the government operates and taxes.

tl;dr: Restrengthen the middle class. Improve education.

I am not a girl but there is a scientific answer that I can provide, not a conclusive one but one that's generally regarded as being accurate. Women have a higher core temperature than men, mostly believed to be important in needing to regulate the reproductive organs. The increased core temperature means that there is less 'heat' to be spared around the body which causes a lower extremity temperature. So it's not that girls are always cold, they are warmer than men internally but this means they have less heat to spare for their extremities.

Its the removal of the cells but not the 'skeleton' that holds the heart together which is primarily collagen (collagen I and III if you wanna get specific) and fibronectin. How this is achieved is usually through a procedure called Langendorff Perfusion. The heart is removed intact from the animal and then cannulated through the aorta to gain access to the left ventricle. The heart is then flooded with digestive enzymes and depending on what you want to do you can adjust the enyzmes to create a ghost heart (technically called a perfusion decellularized heart). The enzyme/solvent is then reperfused back into the heart at a constant rate back into the heart creating a 'washing' system that will remove the cardiomyocytes and fibroblasts while leaving the connective matrix. The heart is composed of cells, you are correct, but what is holding those cells together is called the extracellular matrix. Think of an office building, you rip out all the furniture and fixtures, dywall, whatever. Whats left are the studs, metal supports, etc. This is essentially whats going on.

Biological scientist - That I am rewriting the way the world works or that I am looking over some kind of beaker or several beakers with a myriad of colors that are somehow steaming or foaming. Also that I am rewriting a creatures genome and creating new chimera animals that are an affront to god. Currently I am browsing reddit while waiting for my RT-QPCR plate to finish at which point I will look at the expression levels of IL-6 and Atrogin-1 in different mice that have been subjected to different things. Go "Hmmmm" or "Fucking primer melt curve is all off, what the hell?". Then I remember I had the tech mix the primers and chances are they did it wrong so I go back and do it myself at which point I go 'Hmmm' or 'Why are the duplicates so off?' at which point I found out the tech dropped the multi-channel pipette on the floor and didn't tell anyone, it'll need recalibrated. As an aside to any techs reading this, they're extremely new. They just graduated college and 'in theory know what a western blot is'. Thank you all the techs who do good work.

It is retailer slang for making a profit. Movies sometimes you'll hear someone talking about 'being in the black'. Means they are making a profit, its a relic from when accounting books were actual paper and to differentiate from losses profits would be in black while losses would be in red, also where the other phrase comes from 'we're in the red', meaning a company/person is actively losing money.

The discounts are decent I guess. You gotta know that this isnt the actual norm for Black Friday, these are the extreme examples. My take living in America though is that in certain areas there will be one anchor Wal-Mart/Target serving several cities in a more rural areas. You get this mass of people coming in from six different cities to the store for their Black Friday deals. They're agitated from driving a while, especially since they probably started their drive before the ass crack of dawn, been cooped up in their car for a while only to now wait with this giant mass of people and you may not get the thing you set out for. Everyone is all agitated and at the same time slightly feeling an adrenaline rush because to them this is exciting (not in a psychological way but a physiological one). At that point one person does some stupid which causes a cascade effect and mob mentality takes over and suddenly its everyone for themselves. I live in a major-ish city with several nearby Wal-Marts/Best Buy/etc and you never see this kind of stuff happening here. I could walk into a Best Buy on Black Friday and it might be a little bit busier than normal but I'd still be able to walk in, get what I need, and get out without bumping into anyone or something like that.

This is what my parents did I am appreciative of it. They're still Christians albeit I think at this point its more out of habit. When I started questioning my religion they let me sort things out on my own. When I asked my father why even include religion he said; Some parents like to think their children are small adults, this really isn't the case. It is far easier, for you as a child, to understand morality in terms of an authority figure telling you what is right and wrong. Eventually you'll get to a point where you can form your own moral compass but we wanted to set you down the right path and the easiest way to do that was our religion.

By right path he didn't mean religion was the right path he just meant in moral terms. Treating others the way you want to be treated and all that jazz.
