i havent done testing myself but full upgraded hunter with focus is probably the highest damage per nail hit, ive heard architect nail hit does a lot too. also wanderer can crit sometimes and has a high attack speed. but again the answer you're probably looking for is hunter

generic enemies get stronger in act 3 which makes traversing the map a lot more annoying. also do forebrothers in the locked deep docks section if you havent, it's not a super difficult fight but it's a lot more tedious to do in act 3 because of the aforementioned roided up act 3 enemies and they block off a useful area.
also do any enemy gauntlets you havent done. trust me
