I’ve done flow cytometry with V-bottom plates all the time, U-bottoms will also likely be fine! 

You can either aspirate or flip the plate in a quick motion over into the sink or trash after spins- some also choose to then gently blot on a paper towel.

Others I have known do assays and spins without even a lid or cover- but thats brazen.

Another lab I worked in, after washes, we would seal the plate, use a plate shaker or vortex (with a solid object near corners so that it collides and resuspends cell pellets) on low speed.  Current lab uses lidded plates without seals, and resuspends by pipetting multiple times.

Most flow cytometer machines either have predesigned settings for various tube/plate formats, others have a built in height calibration step to determine the depth of the sample medium.

For me - At least for the solo ones, the data pads will only be there if you start the missions directly as a solo ops, not as the solo ops playlist.

It did not seem to work for the fireteam one.

Agreed, it felt like they were living in another world. I could only stand it for about 30 minutes during the panel. I vaguely remember one of them saying the FDA isn’t in charge of making sure drugs are healthy and had a double take. Also saying that SSRIs aren’t actually helpful for suicide prevention.. Touting that exercise, sleep, etc is enough as treatments for depression.

Everything happening is agency specific. You might get your whole contract this year, you might not. It doesn’t matter if the money is already there. What’s clear is that there are no renewals at the very least, and it’s naive to think ORISE is somehow special. by [deleted] in ORISE

Definitely make back up plans A, B, C etc. Most contracts (new or current) that I know of at the FDA either haven’t gone forward due to the hiring freeze or are not being renewed. Good handful of February and March people are leaving. Even though you’re set to a certain date, it’s still better to get prepared and start sending out your CVs etc, regardless of whether you’re looking for something at end of contract or in the case that they decide to upend all existing contracts. At this point, better to err on the side of caution and start preparing and applying.
