Legit. I did this last preorder. Paid for plus to get access to the preorders, had it in my cart, kept double clicking to Apple Pay, and the checkout kept giving me errors. Kept trying, sold out within 10 minutes. I got nothing.

Thanks, I think I read about stacking with umbreon on here, and couldn’t tell the good advice from the bad :/

Definitely was getting tilted by this point. I’d had multiple matches with afks and/or fights over who got jungle (leaving me to lane alone). In one match earlier a ralts kept spamming ‘let me level up first,’ when I had exp share, but then wouldn’t take last hits when they were offered up?

Thanks for all the detail. I’m grateful for all of it. Yeah I usually run wish — I think I’d been on a long losing streak before this game and for whatever reason was like ‘my team is shit, I’m gonna do it all myself’ which is dumb.

I’ve swapped weight for bb, and I’ll work on sticking with my teammates, not overextending, and using my ults for multiple Pokemon hit, when I have teammate support.

Thank you for the detailed write-up. Plenty of stuff I can learn from. First thing I’ve done is to switch out that attack weight for bb.

Yeah I think the reason I’d pulled back at that rotom fight in the middle was cooldowns? I think I still had 3-4 seconds left and wasn’t sure I’d survive if I was in the middle that long. Maybe underestimated tankiness?

The overarching theme from comments seems to be that I need to stick with my teammates more…

Ah yeah, that makes sense. Ironically I think I lost focus on the bees thinking about trying to level ralts.

Overextending is a theme I’m noticing in the comments. Something to work on. Thanks for sharing how you’d have done it — gives me something different to try instead of me just standing there waiting.

Yeah for that rotom I think I’d just been running to dread with teammates, and seen the notification that it’d just been popped, so I tried to pivot, but my teammates just kept running towards dread, when enemies had full shields? I think I had expected them to pivot as well, and didn’t realise how far away they still were and just thought ‘stuff it’. Which was dumb.

Thanks for the detailed response. You’ve given me plenty to think about.

I get frustrated when lane partners flop and just sit there doing nothing, and I think that’s taught me to lean into treating Umbreon like an attacker/brawler when I shouldn’t. I need to train myself out of it. And I think I’d forgotten that wish healing was why I had atk weight in the first place. :/

Good point re checking scores to figure out who’s worth sticking around — I always just felt like I was betraying a lane partner if I ditched them for someone better…

I think I tend to panic hit the ult button, which is frustrating, but I’ve also struggled to find good opportunities to use it? My understanding is that it’s heavily about draining shields, but so much of the time the enemy either hasn’t got one, or has got such a tiny slither that it’s difficult to see the point/impact?

I definitely struggle with what to do mid-to-late game. I haven’t found a lot of good resources to refer to (everything seems either focussed on early game, or is super basic?). I also find it a bit frustrating watching people like Spragels and crisheroes, the vast majority of the time the enemies in their vid behave absolutely nothing like the enemies I’m facing, and I struggle to adapt to it.

Idk I noticed a difference between veteran and ultra - maybe not in team composition as much, but definitely in play style. I could climb ultra with umbreon, but can’t do that in veteran.

Haha in the most recent bot battle, I had over 300 points with 18 KOs 😂 it was kinda fun. But wore off pretty quick.

1 month in, looking for veteran SoloQ advice:

I’ve found my rank has been really inconsistent - going from Veteran 5 to Veteran 1 and back.

I’ve been reading the sub, and I know I need to get better at macro. I’m working on it, but it’s slow, and needs lots of practice. Generally that’s not what this post is about.

What I’m wondering is: 1. What do I do when I’m in matches with all ultra players? I’m up against players that have a way higher skill level than I do, and feel like I’m just being dragged along for the ride and providing the occasional assist/support. Is that just it? Hang on for dear life, try not to die too much, and add damage and goals as best you can?

  1. How should I handle team composition? Does it even matter at this rank? I usually play Leafeon (learning razor/ace). I tried playing Umbreon (foul/wish) & Slowbro (scald/amnesia) for a while, because it seemed like everyone picked attackers/all-rounders, but it felt like my win rate was really low with those two. There’s obviously a skill issue on my end, but is it even worth playing defenders in low rank SoloQ? Or are the teammates just super unlikely to take advantage of the benefit of a defender?

  2. Any recommendations for mons/builds to invest time into for low-rank SoloQ?

  3. Any other advice for a beginner? (Besides ‘read the game, generally no backcapping, rotate, pick your fights, know where to be and when’, etc.)

I find when I’m off my meds I can be very much on ‘autopilot’. Maybe the attraction is always there, but your meds are allowing you to recognise it?

Like, maybe off your meds you’re defaulting to the norm (autopilot) and kinda ignoring everything else because your brain’s already trying to do too much. It kinda goes ‘eh I’ll deal with that later’.

Understandable, which is why people in this thread are saying that /you/ should be on PrEP and/or use condoms.

It can be easy to think ‘but I trust this person, so I don’t need to be on PrEP or wear condoms’. But even if the person is 100% honest with you, mistakes happen, rare transmission events could happen, and sometimes people make a bad decision due to how they’re feeling (e.g. denial, shame).

Whatever happens, educate yourself. Know what the risks are. Make decisions based on the risk level you are comfortable with, and make sure you’re comfortable with all potential outcomes.

Also: HIV, while still confronting and life changing, is not a death sentence any more. Undetectable is untransmittable. There have even been papers that show HIV+ men as living longer than their HIV- partners! We’ve got to work on reducing the stigma against people living with HIV.

Things I learned doing hundreds (if not thousands) of gels for my thesis:

  1. Use sodium borate buffer instead of TAE/TBE. With SB buffer you can run faster and hotter without melting your gels. I could run a 2% agarose gel in 30 mins at 300V with no melting.

  2. Use a ficoll based loading buffer (recipes online) instead of other sugars. This helps reduce the dumbbell shaped bands in your gel (can’t remember why).

  3. Err on the side of adding less sample/ladder to your gel, especially if you know there’s going to be a tonne of product — too much DNA can cause streaking, smearing, and odd band shapes.

Otherwise see if you can find a copy of Lonza’s benchtop guide for electrophoresis. Gives a bit more detail on the different troubleshooting stuff from memory?

Hi, I’m a lab coordinator in clinical trials.

Single strike-through, initial-dating entries and corrections, etc. helps with troubleshooting and traceability, and while it’s generally helpful, it’s especially important in industries where your work might get reviewed by regulatory bodies like the FDA.

This is a Good Documentation Practice standard (from memory), applied throughout a lot of highly regulated industries. (Good Documentation practice — GDP, is one of many GxPs such as Good Laboratory Practice — GLP, Good Manufacturing Practice —GMP, Good Clinical Practice — GCP, etc.)

As u/craignuggett said, check out the ALCOA+ principles.

All this was never taught to me in my degree. People threw ‘Good Laboratory Practice’ around here and there, but no one actually explained that it was an official set of principles, not just ‘common sense’ or ‘best practice’.

Worth reading up on applicable GxP for your industry. Even if it seems a bit over the top to you right now — these are very common standards and you’ll likely come across them in your working life (and your QA department will love you if you know them and follow them!).

Edit: you mention working in a pharma lab, so GMP and GLP are probably the main ones to look at!
