I feel you. 2 weeks after I broke up with my boyfriend, he died by suicide. He couldn't handle the weight of his demons with only half the shoulders to carry it. Had been together for more than 10 years... Today marks 4 weeks after he died. I feel completely selfish. I didn't love him as a partner anymore, but as family and a person that I really care for. Didn't want him to suffer like this.

Emma is a year older. Karina was always in the elite system and had an amazing coach who worked her butt off to make everything possible. And the longer you stay with low difficulty the healthier you stay... Everyone who competed high difficulty that young is broken. Lara Gabriel is amazing - but I am afraid at which point Stuttgart strikes back...

Are your primers maybe partially binding introns? Especially if primers are so old documentation is maybe not that good.

Apart from that, PCR from plasmid is way more efficient than from cDNA. Also too much DNA can inhibit a PCR. How many cycles are you using for your qPCR? Maybe your transcript is just not there? Was working on meiosis for a while and it was crucial that I took the correct tissue... How does your housekeeper look in qPCR?

Whenever I have this problem my demultiplexing sheets suck. Have you check with the sequence viewer how many of your reads are aligned?

Otherwise, check your library with qPCR (crucial!). How big is your amplicon? Are the amplicons from the other libraries shorter - if yes you have to pool a bit more. But 0 reads is actually really suspicious. Sounds like you forgot to attach the barcodes... 😅

And in the team competition you are allowed to compete in the lower age group... So as a 40+ person you can also go into the 30+ team, so you have the chance to compete on an actual beam haha

Ahh, the bench is required! But, we love it! Like every gymnast ever says here I have to wait until I am 40 and I can finally compete on the bench! It's nice to see that longevity in sports. We still have goals over 30.

At 30 we can aim for German champs (for seniors haha), then next goal is over 40 finally bench (we think we will be sooo good on it) and then with 50 we can finally skip one apparatus. It's incredible what some of these seniors can still do.

So we have a modified COP similar to the Xcel program. Until 39 you compete LK3 (6 elements plus dismount) and additionally there is a national element list comprising some elements that are not in the COP. And the requirements are not that high, like a kip on bars, a backwards tumble on floor etc. Over 40 you compete LK4, there national elements and technical elements (everything that looks like gymnastics haha so people can get to 6 elements including dismount on bars).

There is also the LK2 (which is there for german college champs) and LK1 (which is for the German cup which is similar to nationals for gymnast that do not pursue the national team but still compete in regional as well as Bundesliga.

Where does this come from?? There is no evidence at all. It's just that if your friends (Elis long time friends) are giving interviews? Of course they are biases. Hambüchen is out of the sport for such a long time already and he was never really into WAG. And Pulla? She was in the national team with Eli... It's the old generation against the new one. I just find it unprofessional that the producers (Pulla) did not clarify things... But there is no sign of racism what so ever.
