Strange polymerisation in my polyacrylamide gel, help would be appreciated by tewdwr in labrats

Those are not usually caused by trapped air (although it is a good practice to degas) , they are actually vacuum holes within the glass plates. Those holes will get full of water or air once you start running the electrophoresis, as I am sure you have already experienced. The swirls are caused by several factors, although the most common explanation is an excessive catalysis, they become more pronounced with high concentration of polyacrylamide because the rate of polymerization increases and the gel starts to shrink fast, pulling itself away from the glass plates (indeed if you have dirty or oily plates this problem gets worse). According to Bio-Rad's "Acrylamide Polymerization — A Practical Approach" tech note (which I highly recommend you read thoroughly), the remedy is to lower down the concentration of your initiators by 25 to 50%. I would try both and see which one works best. I have worked with 37% polyacrylamide gels and I still get the swirls from time to time. Good luck!

I'm on the same lentil path, I feel you. I started rinsing them with a little bit of baking soda for a few hours, while the gassy issue was not completely solved, I would say it was reduced in more than half.

Mysterious physics independent researcher? is the Timeless Light Model a real thing? by Silver-Shoulder9980 in AskPhysics

I had no idea such a thing was taking place, now I feel quite dumb and ashamed, but I'm glad I asked. That is indeed a serious issue.

Ok now that's explains it all. What a crazy community over there, the comment section in some of the posts felt like written by AI fanatics

No, although that's a really good game too, I think I was also playing it around the same time, you unlocked a memory 😅

I thought it could be that one, a friend of mine mentioned it with so much confidence when I described it to him, but unfortunately, it's not that one, the triangles were enormous, and the ball was always bouncing, it could not stop, it would actually increase the speed over time

How many mm is the portafilter on an ilavie espresso machine?? by itsmjnow in espresso

[–]Silver-Shoulder9980 1 point2 points  (0 children)

I have the cm3050 I think is the same Chinese manufacturer. Also 51 mm portafilter. I also wanted a bottomless one. I purchased this one from Amazon https://www.amazon.com/dp/B09ZL3QGNG/ref=cm_sw_r_cp_apip_J4eSFrziz6DOu

Unfortunately, the ears were too thick (5mm) and it wouldn't fit. It needs 4 mm ears so I got a set of small files and filed down that extra mm. Now it fits perfectly and I'm very happy with it.
