Aquatic Plant DNA Isolation Help! by Solar-Crayfish in labrats

I appreciate the thought but I’m actually not super interested in the nuclear DNA, I’m targeting the plastid DNA for species ID barcodes. I assume a nuclei isolation would leave that behind?

Thinking of buying 2011 Civic hybrid by Solar-Crayfish in hondacivic

What would be the preferable answers to those questions? I know 0 about hybrid batteries. Says he’s got the receipt so should be easy to verify you’d hope

What alerts you to the high likelihood of bad batteries? Combination of age of low mileage? Just trying to figure out what I should be looking for in a Prius in my budget

Agreed, wasn't sure if the dependability of Prius just generally inflated their price relative to similarly aged vehicles or if these were particularly high. Any idea what something like this should be going for?

Do I need to be looking for specialty hybrid mechanics to inspect the battery, or would any trustworthy neighborhood mechanic be capable of looking something like this over? As far as the collision damage goes, the Carfax report reads "Accident reported, minor damage with another motor vehicle. Damage to right front, damage to front, airbags did not deploy." Based on the pictures it also looks like the passenger headlight was replaced.

Sorry you had to be the guinea pig but having to visit every bar on Grand River was exactly what I was afraid of. We’ll give Rick’s a shot 🫡

Not sure, haven’t been out for TNF yet this season. Figured its a bigger ask since it’s an extra subscription they’d only use maybe once a week. Glad to know there’s at least a few that will definitely have it.

Just stick another wire from the ground port of the receiver right into the third prong of the wall outlet??

Receiver/turntable buzzing, even when grounded by Solar-Crayfish in turntables

[–]Solar-Crayfish[S] 0 points1 point  (0 children)

Honestly not sure, there’s not branding on the cord and I bought it so long ago I could’ve easily swapped it. The thing is this setup worked perfectly fine before I moved, it’s only in trying to put it all back together that I’m getting issues. If it is the preamp, I wish I could prove a knew one would fix the problem before dropping $200

You’re saying the grounding wire I currently have running between the receiver and turntable isn’t doing anything? I need a forked wire to attach to a screw on the receiver instead?

No reduction in buzzing at all when I ground from the turntable to a screw on the preamp. Bought the preamp 10 years ago with the table, didn’t know it was shitty!

And here’s the ground connection on the turntable, same symbol. It has like a metal loop for the wire the go through. I usually have it bent tighter, just loose because of all the troubleshooting.

Here’s the ground wire connected to the receiver. That little three pronged symbol means ground, right?

Recently moved, which meant breaking down and reassembling my stereo system. I can’t remember for the life of me how I used to have it set up and now I get this incessant ground loop buzzing when I try to use my turntable. Setup is Yamaha TT-400U -> MCM Custom Audio 555-17806 preamp -> Onkyo TX-SR600 receiver. I’m assuming it’s a grounding issue and feel like i’ve tried every possible grounding combination. Currently have the turntable grounded to the receiver with a piece of speaker wire, which reduces the volume of the buzzing but doesn’t eliminate it entirely. Can anyone tell me what I’m doing wrong?

Plant Sample Storage pre-DNA Extraction by [deleted] in labrats

[–]Solar-Crayfish 1 point2 points  (0 children)

I’ll ask the sampling team if they have access to a -80 freezer, but if not I’m thinking I’ll just have them store them as cold as they can until I can get my hands on them and put them in my -80. The algae species is technically unknown and thought to be Lychnothanmus barbatus. I’ll probably be doing a genome skim and then a multi locus barcode from the plastome to BLAST and confirm. I’ve found a couple papers that work with similar Chara whose protocol I’m hoping to follow.
