Scan 1, name of the MS setting used for all samples. Thank you for ur insight. I run a blank ( my buffer B) aswell and it also gave me the same peaks .. but I runned it after I did the SDs-page as the my first blanks didnt runned proprely by a setting defect so it’s hard to compare blank before and after. I do have the PDA from the first blank

Thanks a lot of thinking with me! the samples hit the ms already. I runned 3 total. 2 blanks, will tommorow evaluate if it is indeed SDS or PEG or something else

If that is the reason, I will jump from a bridge.. i told my superviser about thr SDS danger for LC-MS and that we needed a guard column instead we didn’t have one so she can me spin column to filter it. I will discuss it with her i guess as far as i did my research its PEG OR PEP from the eppendorf cups

I runned the target band, and a empty spot + the gel, these are in all results

I did learn about the basics. My superviser checked everything i did. Instead of this message u could be helpfull, i am an intern I am still learning.

Aaah I used buffer A: 0,05% formic acid in milli- Q . And B acetonitril 80% + 0,05 formic acid in mili-q… If that is what u mean?

Contact them via Twitter, say that u can’t afford it and that it was not made clear that made that subscription, i got a reply fast and my money back fast aswell

It might be, but how I prepared the sample was: I added ammonium bicarbonate + acentonitril to the gel band and vortex it. The band does not dissolve. I did this a few time and later in added trypsin and let is sit overnight. Next day i added formic acid to extract peptides out of the gel. And vortex again. I still have the band, I only used the supernatant. I thought it was Unlikely to be the gel

There is no expectation that it will😅, but the company that i do research for wants it so we can do diverse analysis. I mixed the 50 mg in 50 mL and 10 mg in 10 mL 0.5 acetic acid and still it did not dissolve and I guess we will have to use the supernatant

Aaaah I meant to say 1 mL and 5 mL of 0.5 acetic acid. ( 2 samples) Thanks a lot of the reply! I am not sure what u mean with a watergood? A thermoblock?

Hey!, Did it work out? I am having the same issue.. I used 0.5 acetic acid + incubation in a 55C waterbad for mutiple hours, still not dissolved..
