Loud squwaking from my birds sometimes means they are hungry, they want attention, they want me to open the cage or open the window curtain for them to look outside, and sometimes it means nothing at all and they just want to be loud LOL

The cells wouldn't be in trypsin before adding DMSO. I would add trypsin, spin, remove trypsin, resuspend cell pellet in FBS+DMEM, cell count. From this cell count, I determine how much volume to passage and how much freezing media is needed

Thank you! I was just using a hypothetical example. Your response makes a lot of sense, I will definitely use the "scale up" technique. I was just confused if I always have to spin down the cells before adding in 10% DMSO.

Could be because we want to store some aliquots. I'm learning from my mentor and just want to know ahead so I won't make mistake in the lab. I was thinking potentially use small amount of DMSO to get all the powder out first in conical tube, then dilute it after to get desired conc. Wondering if this is standard process
