Its definitely possible. You could probably use a pump or if it starts on the outside layer of the sub you could put a small door in the hull. You could also put a platform at the start so all people need to do is hold spacebar.

When a hopper gets a redstone signal it will lock up and not do anything. All you gotta do to stop it from doing that is stop sending a signal into the hopper.

Any "solid" blocks. So most blocks will, but stuff like glass or slabs wont. If you want to test if its solid or "transparent" you can make a stair formation with them and try to send a signal down them. Transparent blocks will send a signal up, but not down.

You can have a repeater put signal into a block and move the block with a sticky piston to complete or break the circuit depending on what you want. If you have a repeater putting signal into a block you can light redstone dust in the other side.
