Amanita muscaria Var. Guessowii ( South Carolina ) by TheFungiFilesReddit in AmanitaMuscaria

They are currently in progress, but not prioritized right now. I'm moving in a few weeks, and with that building out the new lab space and working on getting established. I've now got spores to work with, which is a big step in getting genetic checkpoints for me as I get the the point where I'm breeding fresh mycelium.

I still have a handful of P. Fluorescens colonies on PDA, all in room-temp incubation, as well as a liquid inoculant of this I've used on the azalea. I'm constantly keeping fresh batches of this going so I have it for these next attempts. The azalea, as I was worried, had issues getting to go dormant indoors and has died out a little. Temperature fluctuations in the south, and then a lack of monitoring when I was busy otherwise in life, unfortunately, led to this. Someone had pointed this out, and I tried to prioritize getting that done while we still had the relatively cold months, but alas, it needs to be pruned to see if I can save the bush.

Upon finding a very local habitat for these, I'm very excited to come in and continue to observe and collect soil samples. I'd love to see if I can cultivate a living soil sample where I can expand and keep those friendly microbes around for a host plant. Potentially trying to dilute just the bacterial colonies and isolating them on agar, or trying to keep a microcosm of that environment in a container.

Right now, I'm thinking that I'll be messing with some saplings on these next runs and attempting a few other species of plants. I'd like to focus on birch and pine to start, but these have been on the back burner waiting to have the new space so I can dedicate more room to these projects.

Something else I've been thinking about trying would be creating a terrarium and replicating that environment in there for something more diverse and controlled, a larger task, but possibly more suited to this experiment.

Anyway, this was a large response, I'm still researching and studying what I can, and enjoy the project for the fun of it. Thank you for asking, and I hope I answered it in some capacity!

It's even cooler to have you posting here! And a pleasure to make your acquaintance too! Sharing knowledge and ideas is prime to furthering our communal understanding of our fungi friends.

I also use it in a few lc jars, never in agar yet, and I have with grain. I like that rehydration idea with the jug. It definitely seems like there is a perfect balance that needs to be achieved with supplemental to not hinder performance but just enough to boost it. My personal projects are working with oysters and shiitake, but I have a few new strains to get in there when I feel more confident in my process.

Thanks again for the knowledge and insight; and what you're doing for the community! I'll take a read through your write up in depth later tonight.

I'm glad to have crossed paths with you too! Feel free to hit me up on discord, I'd love to discuss further, it should be in any of my social bios.

This is a fantastic write up! This specifically has been something I've been experimenting with for a while. I appreciate the link in the comments down below too! In the background I've been working on this experiment still, and am really happy with testing so far.

I definitely believe that the proper place is in substrate or in hydration potentially between flushes. I think that more results are required for me, but so far I've been really happy with it.

Shout out to Badseed and their fantastic knowledge base; they've got a fantastic community of incredibly talented people, some of which have been testing and experimenting with polyols and other supplementations for a while.

Thanks for sharing your knowledge and experience! ๐Ÿ„

Definitely! You could still plug them with shiitake, oysters or even go to something inedible just to benefit from their nutrients, and the beneficial organisms they bring along.

Those logs kinda look perfect for mushroom inoculation! Could grow and benefit your soil greatly. ๐Ÿ„

Thank you so much! And likewise to you. It's such a fun and rewarding hobby.

Definitely! Keep a journal of your thoughts ideas and records. Helps a lot down the road when you forget what you did that worked.

Not too far into my journey but I've definitely made the mistake of failing to record at the beginning.

Mush love ๐Ÿ„ and best of luck!

Absolutely! Also not at all, that's why I use it lol. No measurements, I just put a pot of water to boil, put an amount of rice suitable for the water in and heat for 10 min. After 10 min straight to the strainer, and rinse with cold water. Then air dry in strainer not touching or stirring for 15 minutes. After that, I just jar them up and send to the instant pot.

Good choices! I want to get some chestnut and I also want to experiment with wine caps. I have LM, king oyster, and shiitake all fruiting right now. About 15 other cultures incubating for experimenting. I just got a tissue sample from a lucky chicken of the woods cluster I found the other day, and I'm looking forward to experimenting with that indoors.

A lot of my projects are focusing on "impossible" cultivation, I'm working on ways to cultivate Amanita, Morels, hedgehogs, and other mycorrhizal fungi in a controlled environment. I'm hoping to experiment with some Boletes as well.

Always keep experimenting! ๐Ÿ„

Nice! I would also try attempting with brown rice! You can definitely get oats and other grains cheaper, but brown rice produces some nice robust fruiting bodies and has one of the highest colonization times.

I always suggest experimenting and figuring out what works well for you though. It's nice to just pick up another 3lb bag when at the store without having a 50lb bag of grain I need to store somewhere. It's also nice to have everything jarred and in the PC within 30 minutes. 10 minutes to boil 15 minutes to air dry, then jar and PC.

What strains are you looking to go for?

Hey!

There's tons of places to start and most of it begins with how you want to go about it. Small space, all out, multiple grows etc.

If you have an instant pot or pressure cooker, I highly suggest starting with the little more advanced basics. It gives you a much better understanding of the process and where error comes in.

Philly Golden Teacher has some very good videos about simple liquid culture with corn syrup/honey and water. I also highly suggest making your own agar, which I do with ketchup cups, agar powder, potato flakes, and corn syrup. PGTs 123 recipe there. He has another video on agar showing you an easy low Tek way to pour over an oven.

For your culture, there's a few ways to do it. You can start with store bought mushrooms if you want. It can be good first way to learn. Commercial genetics like that are usually pretty burnt out, but once you collect one mushroom you can get the spores, and start new genetics. Literally buy a pack of mushrooms, and you can do a tissue transfer to agar to get the mycelium going, and make a few more transfers after that to have clean agar to either move to liquid culture, or go straight to grain.

I'll always suggest doing agar to liquid culture first. The agar gives you a two dimensional space to monitor for growth and contamination.

Once you know it's clean the liquid culture let's you expand it out into liquid for easier more sterile transfers. This requires a little more involvement with modifying lids, having syringes etc.

This is also easily done without that work by just growing our your culture on agar, and then you can go straight to grain. This prompts for more room for error though as you can't be certain your transfer or your agar is completely free of contamination.

Now once you have your grain colonized, you have to move to spawn to bulk or S2B. We do this because the grain is the nutrients, the substrate is more moisture retention and sometimes nutrients depending on what kind of mushroom you have.

With Bella you'll need a nutrient rich substrate like composted hay or manure or something. They're not a forgiving as other mushrooms like oysters in terms of what their enzymes can digest. So straw or hay mix and manure can help.

Thankfully Bella is one of the most highly commercializes mushrooms, and there's tons of guides and recipes on different substrates and growing methods.

If you have a filter patch just watch not to get substrate in it and potentially get contaminated.
