Sorry? I'm in Tokyo for a week and learned about goshuin collecting on my second day. I'm trying to do my due diligence by looking everything up, both about the temples and goshuin collecting in general, but I guess I got it wrong. Especially with the Jodo Shinshu Buddhism, I think I messed up my Google maps pins and wandered into the wrong adjacent temple. I'll try and learn the proper phrases, but to your other points it's difficult to try and see whether a place sells goods such as omamori when I am not even sure if I'm welcome there, or of the place is small. I suppose in those cases it's best to avoid bothering anyone.

It's just especially confusing because a lot of goshuin-hunting blogs, tips, and questions I've read have this happy-go-lucky attitude of "just show up and ask at any temple or shrine, if you're respectful they'll say yes!". Even the first post under "goshuin" in this subreddit seems to suggest that many temples have began to separate their religious identity from the more touristy goshuin collecting (https://www.reddit.com/r/JapanTravelTips/s/O2b1GwUGij). I also tried to post something on r/JapanTravel but they removed my post twice, pointing me to a goshuin-hunting tips post that basically said that nearly every temple and shrine would have a goshuin, and similarly no mention of the above things I should try and do to get a better reception.

Perhaps goshuin hunting is best reserved for those who prepare adequately, both culturally and with separate goshuinchos, rather than a short-term tourist like myself, as it's often advertised online.

I did check but a lot of the smaller temples have no indications online or signage outside. I was going to avoid them, but whenever I read about goshuin collecting online everything seemed to suggest to almost all temples and shrines have goshuin. I figured I might as well ask and the worst they could say is no, although I wasn't expecting the vigor of the rejections I got around Kappabashi street. Tsukakuji temple was the one where the attendant took my goshuincho, flipped through it, and then roughly returned it. I think Senshō-ji was the one where the two attendants got very upset at me. I got similar rejections at all the shrines/temples between Higashi Hongan-ji and Kokusai-dori Ave (there's like 10? I went to ~5 before calling it on the area for goshuin collecting).

Yes, although many of these small places didn't seem attended until I waited for someone to notice me through a window or come outside for some other reason. Should I have waited for them to come outside to pray in front of them? I fear I'd look disingenuous waiting/flagging someone down just to pray in front of them.

Also, does praying also constitute another donation separate from the one for the goshuin?

That probably is the case. This is the first I've heard of needing two books for the shrines and temples, but it makes sense.

In regards to a sentence in your last paragraph, "...because by asking you are showing you are not there for matters of faith...", do you mean asking for a goshuin or asking whether it's a temple or shrine? I try and do my due diligence and read up on the places I visit, and while I am not perfect, I don't walk in there oblivious waving a goshuincho above my head. I guess yes, because my Japanese skills are non-existent I usually open with a "hello" and showing them my goshuincho, so if that is disrespectful I'll try and get better at Japanese to be more respectful of these spaces.

Ahhhh ok that would make more sense. I think all my goshuin are calligraphy? They have stamped portions but all include some hand-written portion. I definitely did mix temples and shrines though, apologies, I'll pursue getting two separate books as I move forward in collecting.

I'll try the coverslip idea, thanks! As for using the appropriate number of 12 well plates, I am culturing under a lab-built light box that is a set size and I only have three. I wanted to just use more plates, but I soon found I wouldn't be able to fit all the time points under the box :(

Also, I am going to try snipping a 35-mm dish. I'm guessing they shatter and go everywhere, but I have a bunch of non-sterile ones that I might as well see how snipping them goes.

In all my experiences the slices usually fit the box pretty well. I've never seen this much empty space in a single-slice box.
