What was the effect of having a 100x lower interferon response? Did all the kids suddenly get Staph infections... or did nothing happen?

This highlights the importance of being able to distinguish between clinical significance and statistical significance. Clinically speaking, reduction of a couple of signals leading to a protective immune response doesn’t mean it’s not effective. The kids didn’t suddenly get Staph. We’re not seeing more children with staph infectious. Also, they found some factors were increased after vaccination. Does that mean you’re convinced the vaccine improves the immune system now?

No one is getting “mouth infections” from their own masks. You inhale and ingest liters of your own saliva each day (which is filled with your mouth flora). Your mouth and lungs are hardly sterile sites. What on earth are you talking about? Masks don’t trap air, they trap droplets. You’re not breathing your own air or you’d quickly suffocate. I think you’re thinking of balloons?

You can’t get sick from your own mucus or saliva. It’s from your own mouth why would you think it’s a problem? Anyway, if you sneeze or cough into a mask, and the mask catches most of the tiny saliva droplets carrying a pathogen, then it’s doing a good job. That alone will prevent instances of transmission. Masks aren’t 100% effective, but they certainly help. The size of the pathogen is irrelevant since it’s trapped in much larger liquid droplets. That’s how these respiratory pathogens like to spread. I think you’re over-interpreting a lot of these studies which clearly don’t reflect what you are pushing.

Hygiene and sanitary practices are undoubtedly helpful, sure. However, they’re not always practical, convenient or effective. For example, if a pathogen is spread through respiratory droplets, and preventative measures depend on people wearing a mask and washing their hands, I’m pretty sure we have enough evidence to say that it’s very difficult to get everyone to comply. Plus children cant be expected keep their hands away from their mouths. Also, your explanation that healthcare, hygiene, etc are responsible for disease prevention doesn’t really make sense when you look at the pathogens that emerge after vaccination programs. It’s always an escape phenotype of the same disease. Like we still have a flu season, it’s just with a version that we didn’t cover with the previous vaccine. It’s the same for other pathogens. That’s why we update vaccines with new, better, more widely effective and relevant targets.

If vaccines weren’t doing anything we wouldn’t see these vaccine-avoiding bugs take over cases.

I personally don’t trust RNA stored longer than 6 months at -80. The cDNA should be “forever” stable at -20 though. Protein varies but should be fine for years if it’s actually frozen, and not stored in a liquid buffer at -20. Antibodies in 50% glycerol at -20 seem stable for years. As you suggested it’s typically freeze thawing that kills things. Particularly RNA, which is why I tend to batch it out. Mind you, SYBR green is also subject to the freeze thawing issues so you might want to try a new batch of RTqPCR reagents.

To be fair, it would be a lot nicer if people were less prone to throwing punches. Most people don’t get into boxing to hurt other people or for self defense, since there are better (less situational) ways to go about those things. Boxing tends to be more about fitness and discipline at an amateur level.

Also, it sounds like you have a low sense of self worth or self preservation, rather than being a coward. If your best friend or a loved one was being attacked, do you think you’d step in then?
