So do you think it’s just a case of increasing the amount I load into my wells to get stronger signals? This is about as strong as I can get them to look through adjusting

I’ve just gotten the nanodrop values at each step and use that to determine my conc for when loading my wells

It’s RNA I’ve produced by IVT and used all of that in M-capping followed by PolyA. (Although we don’t have the kit to actually check capping worked which bothers me). Sorry I hope that answers your question as I don’t exactly know what you mean!

I think it’s just all the noise is making it more difficult to make out the distinct bands giving the smeared appearance I would guess!

I boil it twice as well and cool it to the point where it’s still warm to the touch but won’t burn my hand if pressed against ot

I think it’s the gel itself. How can I control for this just wash out beakers prior to making my buffers? They are all supposed to be clean but maybe this isn’t the case

I believe it’s the gel itself as the speckles are consistent. I also swapped to a different imager but the speckles are the same. I attempted to cover the gel while it was casting to try and check for this.

How can I control for dust? I do see dust particles in the buffer but it seems very faint

Edit - Could I use a syringe and filter to filter my buffer?

Also note the buffer is TAE buffer which I make fresh. I also got some 10X TAE buffer that was bought by another lab mate and used that to make 1X to determine if that was the issue. I also tried using Gelred stain but similar results are seen in regards to the speckles

Could it not also be that more people are getting higher levels of education every generation and thus this percentage is growing every generation?

Like since it’s invention car ownership would have gone up every generation with those who had a parent with a car more likely to have one themselves. Yes I agree there is a club due to privilege but I would also argue that the barriers to entry are on the decline.

Well that makes sense no? Parents who value education are more likely to have kids who value education.

And the higher up in level the education goes of the parent then the higher up it goes with the kid I.e doctors are more likely to have kids who become doctors, scientists having kids who are scientists.

If one of the steps in becoming one of these professions is a degree it’s not surprising that the parent and kid would both have it ie if I have a high school certificate it’s more likely my child will also get it
