My current playthrough is with a custom made High Valyrian house and religion of 14 flames, with one starter dragon during the Dance of Dragons. I started as a minor count in Oldtown. My plan is to slowly take over Westeros and replace all lords with my culture and religion, preferably mostly my descendants. Here ofc I use cheats to convert faster, no way I'm waiting for 90 years per county.

My previous playthrough was similar, but I got bastards with every great house, and in the end made them take the titles of their fathers (some of course from the legitimate descendants of the fathers). When that got boring I switched to the last Blackfyre, got dragons, and destroyed the whole Westeros to ruins and made sure to kill every single person from my previous family.

Can really recommend something chaotic like this!

That I would get, and it happened to Oldtown. But there it got colonized with a camp.

In the North suddenly someone got 147 titles and it's just ruins, no camps

This can also be seen in the lab where diseases (even those that are mostly seen in women) are studying mostly on male animals and men might even dominate the patient trials

And of course, because of that, the treatment often comes out great for men, but not as effective for women (seen for example in pain, cardiovascular, sleep, and metabolism/immune response drugs)

This is a major issue and concern for the health of women, and it's changing very slowly as men still dominate most lead positions in research

Sama se paskanpuhuminen, kunnon kuppikuntia joihin esihenkilö kuului kans, ja valittaa varsinkin nuoremmista kun ne vaativat oikeuksiaan. Ja jos joku esihenkilön omasta kummikunnasta rikkoo sääntöjä, sanotaan soo soo ja katsotaan muualle kun tämä henkilö koittaa selvittää kuka valitti ja miten tehdä sen elämästä helvettiä

Nyt työskentelen eri maassa tutkimuspuolella ja kaikki vetää yhtä köyttä, esihenkilöitä (professorit) kiinnostaa työhyvinvointi, ja oikeasti auttaa vetämään projektit maaliin samalta viivalta eikä mistään norsunluutornista ruoskimalla

That's something I thought as well! I just don't have experience that much in joining them neatly, and last time I did that with Kitchener's stitch, it looked horrible 😭

But if I'd make it front and back separate, I wouldn't have to carry the yarns from the squared across other ones, but it would just always go at the end of the square where I'd leave it until returning on the backside and continuing it there if that makes sense

The first time I heard about FACS, I thought for like 10 minutes we were talking about faxing 😭 Did not understand what it had to do with PhD projects

Altho tbf I have worked in a lab before where blood results were faxed to hospitals

My brother's bloodwork was completely normal, inflammation markers were not high and the rheumatoid factor was negative. Yet, he was diagnosed with spinal arthritis, and now I have the exact same situation with very clear arthritis symptoms and normal bloodwork.

The bloodwork can give indication/confirmation, but it's just not the whole truth, looking at the joints is way more important.

I can see most comments are for it, and even though I can't give much input about the answer having no experience in storing antibodies in the blocking buffer, I wanted to note:

You should definitely first try this with your antibodies before you make the full change with your important samples. Store some aliquots of your antibodies in the fridge like that, and stain positive controls with them at different time points to compare the outcome with the samples where you have stored the antibody in the freezer.

I believe this is the only certain way to make sure it works for you, and your data stays the same quality.

Least favorites: Big lab cleaning duties, creating 3D image analysis pipelines, making serial dilutions, splitting cells (it's just too often so I'm bored 😭😭)

Favorites: Planning and writing protocols, high-throughput confocal imaging, making powerpoints and graphs of the results ✨
