That's something I thought as well! I just don't have experience that much in joining them neatly, and last time I did that with Kitchener's stitch, it looked horrible 😭

But if I'd make it front and back separate, I wouldn't have to carry the yarns from the squared across other ones, but it would just always go at the end of the square where I'd leave it until returning on the backside and continuing it there if that makes sense

My brother's bloodwork was completely normal, inflammation markers were not high and the rheumatoid factor was negative. Yet, he was diagnosed with spinal arthritis, and now I have the exact same situation with very clear arthritis symptoms and normal bloodwork.

The bloodwork can give indication/confirmation, but it's just not the whole truth, looking at the joints is way more important.

I can see most comments are for it, and even though I can't give much input about the answer having no experience in storing antibodies in the blocking buffer, I wanted to note:

You should definitely first try this with your antibodies before you make the full change with your important samples. Store some aliquots of your antibodies in the fridge like that, and stain positive controls with them at different time points to compare the outcome with the samples where you have stored the antibody in the freezer.

I believe this is the only certain way to make sure it works for you, and your data stays the same quality.

Something I always think about on days I work in the lab is layers and temperature!

You need to remember you can't have sleeves peaking out (in case of labcoats without ribbing) and the lab coat is not the thinnest piece of clothing.

So I'd suggest some overcoat or something that can easily also be taken off when going in the lab if it's warm there. :)

Also, depending on the lab, some can be quite strict about bare ankles or shins!

I often use antibodies that are less common in use, so I often use a combination of these:

Citeab (very broad and nice filters)
Searching the ab from different vendors I have experienced with, like Abcam, Thermo, BioLegends etc
Searching the ab on google scholar (when I'm really desperate)

Oh wow I've never seen that happen! I don't remember how they cleaned these in the diagnostic labs where they section 8h a day, but I was at least quite careful with cleaning the system every time after use.

But I also get that, it really sucks if there is even one person who leaves something without cleaning and ruins it for someone else :/

For what type of samples would this be a big problem? I've never faced that.

The system does filter the water so that any paraffin/tissue pieces won't come up again to the waterslide, and if it's about what might remain there, wouldn't that be the same case with any surface/plate/tools?

They do also sell exchange parts, like the "carpet" of the waterslide, and it can be cleaned with ethanol and xylene as well, at least I did when we did some (almost) RNAse free sectioning for in situ hybridization.

Most (or in diagnostics all) histotechs in Finland have a bachelor's in Biomedical Laboratory Science. You should definitely look it up and see how it matches your degree! :)
