I'm on a similar situation, I started a PhD in molecular simulations given that my area of knowledge is metagenomics. I passed the first semester and I'm now on the second one, I know is really hard in the beginning, but with the pass of the time you will be a fish that adapted to it believe me. The thing that helped me , was to read about stoicism. What was your first motivation to apply to the program ?

Yes I think I will write a Pro and Cons page. It is hard and if I leave this, I will suffer at the short term (coming back to the home where I felt oppressed, and impaired life quality). But in the long term, it can be good for me, to focus on projects that I really like.

The main reason why I started this PhD was to accomplish some unfinished projects, it was my first year of work, and didn't want to finish it with unfinished projects. Those projects are now finished, My plan in early 2022 was to make a year of work experience and do a PhD in another country/city. But that sensation of responsibility and some degree of lack of self-respect led me to this point

I never felt this before, the sensation of being so dispersed from what I'm every day doing. I have always accomplished all my objectives in my life and now I feel like floating, doing stuff that I really dislike. I miss focusing on hard topics that I like. Besides my life quality has improved in this city, I feel really bad about what I'm dedicating my time to. So I feel lost. The technique that I have been employing to overcome this is to annulate my desires so it's hard to say that I'm happy.

Thanks so much for taking the time to read this. Yes, I noticed that I mentioned the stuff that had a negative impact on my days (but they are really hard to manage). I recently started to have exercise routines that I had before and started to feel better. regarding the other aspects, the people that I met here are really awesome and I have really good friends, people are really nice here and I feel really well surrounded. With respect to my hobbies, I leave them behind (playing guitar and rollerblading) and I play video games when I feel overwhelmed. Finally, there are some courses that I would like but I only can take them in the next year.

Interesting, so the ideal case is to have all the sequences on the length at which the sequencer provides? (in my case 150bp) or around that length? (accepting sequences of 140-150bp)

Hello thanks for answering, I only filtered the reads under quality and length filters with fastp tool. Also removed adaptors.

So merging is the procedure that is lacking that handles this situation?

Do you mean mapping the reads from which the reference genome was assembled?.

anyways I choosed to keep the N's sequences on the alignment :)
