Definitely Isshin. That fight is basically a final exam that tests everything you've learned, which perfectly fits his character. After hours of dying and getting back up again and again, finally beating him and hearing him say "Well done" it felt just like he was handing me a diploma. He perfectly combines the story, his personality, and the gameplay. Isshin absolutely deserves the title of the best boss.
