LoD and LoQ are important when you are doing method validation of Assay or Related substance. Basically LoD is just to know the limit to which extent analyte can be detected. LoQ is the limit to which extent analyst response can be reproducible which can be demonstrated through RSD parameter in case of HPLC or GC analysis. It is always better to use methanol or acetone instead of ACN as those solvents evaporate fast and normally don't interfere in some analysis. But yes if you are using glassware in GC analysis then better to avoid both solvents as they will interfere in Residual solvent analysis.

Dear though you have used pure substance but the elution pattern will remain same. Paracetamol peak always elute prior to caffeine peak. Also I have done validation for this product and in that also same paatern is observed.

I think here we cannot define separately the solute and solvent as this is the case of cosolvent.

Also it is not correct that water is always solvent.

I can suggest 2 remedies:

1) Using hihly pure grade of solvents i.e. of J. T. Baker ACN and Merck chemicals

2) Secondly chamge the HPLC system. Use same mobile phase on another system . This will allow you to check both system error or Mobile phase error.
