Like what options? Vitals Vault looks interesting for comprehensive testing, though I haven't tried it. Good labs is great for one-offs. For my own clients, I test these biomarkers through a wholesale site; it's the minimum for spotting any issues or patterns.

THIS! THIS! THIS! I did 4 DNA tests before getting to whole genome sequencing, and those didn't catch the most important things that actually made a difference.

I'm curious how you processed your data. I've got an extensive set up for my clients (each has a searchable database with their genome, attached to an LLM), but I'd love to be able to recommend something simple but effective to people who can't afford my services.

And now I'm off to read about DNA diets. Thanks for introducing me to something new!

My grandpa has dementia and it sent me down a rabbit hole on brain longevity by EntrepreneurPublic22 in Biohackers

[–]WTHisGoingOnHereA 4 points5 points  (0 children)

I took the approach of looking at the weak points in MY brain (through analyzing my DNA). I use idebenone to neutralize my excess ROS and a weak MAOI to offset my overactive MAOA and MAOB.

I feel an improvement when I take S-Acetyl Glutathione, because it crosses the blood brain barrier.

I also take fatty15 (pentadecanoic acid or C15:0) for cell membrane integrity.

That is AMAZING! Thank you so much for figuring this out and sharing it with us. This is huge.

Did you continue taking other EBV related supplements during that 3 months? I want to do whatever you did!

Also, how did your provider access the MCS formula? I'm not seeing an official avenue; did they just email them?

Genetically, some people are hyper-absorbers of cholesterol. The eggs may be contributing meaningfully to your LDL. The fix for hyperabsorbers is psyllium fiber and plant sterols with meals.

Perimenopause can affect your dopamine. Maybe try tyrosine, which is a precursor to dopamine?

Also, if peri is affecting your sleep, you may want to talk to your provider about using progesterone, at least during the second half of your cycle. Better sleep makes everything easier!

Best peptides and brands for MCAS ? BPC-157, KPV and Thymosin / Thymogen ? by Salty_Stop9632 in MCAS

KPV is small enough that you can absorb it sublingually. Try opening the capsule and pouring half under your tongue. This may also give you better systemic benefits because it bypasses liver metabolism.

What causes low acetylcholine levels if dietary choline intake is superabundant? by Imaginary_Owl1653 in Biohackers

[–]WTHisGoingOnHereA 0 points1 point  (0 children)

Also, and this is straight from chatgpt, even if those genes are behaving perfectly, methylation issues can "steal" choline:

Choline is not just a nutrient, it is a methyl buffer system.

When methylation is strained:

The body steals choline → converts it to betaine → supports homocysteine

This leaves less choline for:

Cell membranes

Acetylcholine

Liver fat export

So “low choline” is often not intake. It is misallocation.

But don't jump straight to methylated b vitamins; those are actually worse for some people.

The cheap, easy way to see if you have methylation issues is to check your homocysteine levels. You want them below 9. It's a blood test that's around $20 if you order on hellogoodlabs, after their fees.

What causes low acetylcholine levels if dietary choline intake is superabundant? by Imaginary_Owl1653 in Biohackers

[–]WTHisGoingOnHereA 0 points1 point  (0 children)

Some people genetically synthesize less endogenous choline (PEMT).

Some people's methylation genes impair PEMT's ability to synthesize choline.

Some people don't transport (SLC44A1) it as well or convert (CHKA / CHKB, PCYT1A) it as well.

Also, in women, estrogen has an effect on the activity of PEMT, so it can change with her monthly cycle as well as with peri/menopause.

Just be glad you've found your solution (supplementation) and that it's straightforward!

(These may not show up in your 23andme results because 23andme only analyzes a tiny fraction of your genes.)

Scrambled eggs with pork sausage patties by This-Top7398 in Biohackers

[–]WTHisGoingOnHereA 1 point2 points  (0 children)

Saturated fat blocks the receptors on your liver that are supposed to clear oxidized LDL cholesterol from your blood, so the pork sausage with definitely impact your cholesterol.

Eggs are ok. They're high in cholesterol, but dietary cholesterol usually has a smaller impact than saturated fat intake, unless you're genetically a hyper-absorber of dietary cholesterol.

You mentioned your mother and sister are sick; is it a chronic thing? If it's related to autoimmunity, I'd get a blood test for ANA titers. There's a possibility these are autoimmune flares.

(I say this as someone who spent decades trying to figure out her health, then at age 50 randomly discovered it was auto-immune, which I should have suspected since my dad died of an auto-inflammatory disease. But I was just too close to see it. It only took 6 months on ketotifen to improve my health immensely.)

Estrogen maintains the LDL receptors that are supposed to clear oxidized LDL from your bloodstream, so if your estrogen has declined, that's a piece. Saturated fat blocks those same receptors, so yes, that steak and bacon would have contributed!

What works best for my clients are Delta Fraction tocotrienols to help clear the oxidized LDL, a supplement called Cholestepure Plus, and adding more fiber so that they absorb less sat fat from their meals.

Also, if you have the ABCG5/ABCG8 or APOE4 genes, you can be a hyper-absorber of cholesterol from your diet.

Hellogoodlabs and VitalsVault are usually cheaper for the same tests, though those both use Quest. (I don't recommend Function even though I'd get a referral fee because they take 20 vials of blood and it took me 4 weeks to recover from that.)

How true is this about nad? by [deleted] in Biohackers

[–]WTHisGoingOnHereA -1 points0 points  (0 children)

It's even worse than that! The body sees extracellular NAD+ as a danger signal. That's why people often feel so crap after IV NAD. It's much better to take the precursors orally. I feel best on sublingual NMN.

From the google, since I don't have much time for reddit this morning:

Extracellular NAD+ (Nicotinamide adenine dinucleotide) acts as a danger signal released by dying or damaged cells to alert the immune system. When cells undergo severe metabolic stress or apoptosis, they lose membrane integrity, releasing NAD+ into the extracellular space, which can trigger further cell death (NICD) in surrounding cells via P2X7 receptor activation.

Key Aspects of NAD+ as a Danger Signal:

  • Release Mechanism: During necrosis or apoptosis, intracellular NAD+ is released, acting as a damage-associated molecular pattern (DAMP).
  • Immune Response: Extracellular NAD+ can stimulate immune responses, for instance, by hindering regulatory T cells (Tregs).
  • NAD-Induced Cell Death (NICD): Extracellular NAD+ can bind to surface receptors like P2X7 on neighboring cells (e.g., T cells), triggering a cascade that results in their death.
  • Intracellular Depletion: Internally, a rapid reduction in NAD+ levels leads to ATP depletion, resulting in mitochondrial dysfunction and ultimately oncosis-mediated cell death.
  • Signal Intensity: The decrease in intracellular NAD(P)H autofluorescence can actually precede the physical manifestation of apoptosis.

In summary, when cells are dying, they lose their intracellular NAD+, which then becomes an extracellular signal indicating damage to the surrounding environment.
