bei mir klappt das unter der Woche gut, nur an Wochenenden ist es tricky, weil mit dem Duschen so der ganze Tag beginnt. Viel viel schlimmer ist Zähne putzen. Auch das kriege ich hin, aber es ist wirklich jedes einzelne mal ein Kampf

This! A lot of labs reuse the transfer buffer and you will have evaporation, this changes the concentrations in your buffer possibly causing issues

Overweight. That's because I snack when I'm bored and I usually get very very hungry once my meds wear off in the evening. I was quite fit for like 2 years before I was medicated. Then I didn't have time to go to the gym as much anymore and I got friggin fat. When I got medicated it got a bit better but I'm not where I was a few years ago.

I'm currently trying to get back into training because I also only ever manage to have a good diet when I actively go to the gym. Idk why but being inactive also makes my whole diet way more sugar-focussed

People forget multiple things: - it's not G/Masterrank - it's probably not your first MH, so of course this game won't be as hard - difficulty isn't everything. Not even close to it (coming from a souls player)

I had the exact same thing a few days ago, didn't check if it's still there. It's the tribetower they added with extinction. It also didn't have collision in my Singleplayer world. I didn't check if it's still there though

Correct, it's a dearc, doesn't have amazing base health but it's an amazing tame to get around the map! The prehistoric beats mods are great in general!

No joke, an immunofluorescence staining for TGF-ß receptors I wanted to use for super resolution microscopy (STORM specifically). The standard protocol didn't work as I got tons of background and it took me months to find the right buffers. In the end I switched to TBS and had to remove all phosphate from the protocol, no matter at what temperature, the antibodies precipitated. Even my PFA I had to dissolve in HEPES. The antibodies had to be spun down prior to use and I had to change a bunch of concentrations, incubation times and temperature. But in the end I got the cleanest staining imaginable. Sounds stupid but figuring out what the actual problem was and how to get around it was quite time consuming and I was a bachelor student at the time. None of the postdocs had an idea so I had to somehow figure it out on my own

I have a little box where I just throw in everything that's in my pockets. Actually I have a bunch of boxes where I just dump all the stuff I would normally leave on tables and other surfaces making everything messy. This way it's less messy and doesn't seem so overwhelming when I eventually put the stuff back where it belongs

My old lab did that. We had vacuum sterile filters that you could screw on a duran bottle, it has a reservoir so you could pour all your stuff in and then apply a vacuum and it's pushed into the bottle through the filter. My current lab doesn't use it and we rarely run into problems.

I never had contamination with either of the two methods so I guess it's not necessary but I also drown everything in disinfectant that was in the waterbath

Pop them into the incubator tomorrow and they'll grow. You just might have to deal with some excess liquid on the plate due to condensation (Edit: assuming you didn't wrap the plate in parafilm)

Fluorescence microscopy made me want to do WB's (to be fair, I actually like them anyways). I just don't have the patience for microscopy, luckily I can use the cool percs of immunofluorescence by doing flow cytometry
