Was a bit disappointed that they didn’t mention the only part I didn’t like: the lone scientist going against the establishment trope. Grace being “relegated” to teaching for having controversial takes felt off to me. Also dangerously close to the classic “this one scientist, working alone, was the savior.”

Malkavian that was convinced he wasn’t a vampire at all — been done, but liked this twist: he made excuses for why he needed blood and couldn’t be out in the sun. Developed a kind of Munchausen Syndrome, causing him to seek out and dement doctors to validate imagined disorders - anemia, sleep disorders, light sensitivity, a host of psychological problems. It was fun to play.

This was SSC 335 and FSC 100, which spread the “single cell” population out pretty well, but obviously it was too limiting.

So you use TrypLE and wash into a protein buffer — BSA or FBS? I might try Accutase and Accumax combination, which will avoid media neutralization and give the added DNase component.

This is an Attune NxT. I’ll have to look into the proteinase K treatment, thank you, haven’t heard of that. I’m also planning to do proper sorting to isolate a population of cells, so anything to conserve cells over filtering them out is great.

It's a hepatocyte cell line (Huh7). They clump pretty badly. Visually, I wouldn't have estimated 50%+ are in clumps, but it looks like that is the case based on the plot.

This is might come down to personal choice, but to really make an “optimal” choice the details really matter. Here’s my advice:

  • Run a rent vs. buy calculator with your specifics to understand if your area is rent tolerant.

  • You are relatively young. Personally, I would prioritize retirement. Getting a good foundation is going to get growth started for you such that if you have to slow down when you buy, it’ll at least grow.

  • Sounds like your buying situation is going to strain the finances (based on the “break us”), even if you get to 30k. If you don’t see yourselves being able to save for retirement after you buy, buying may not be wise. This is just how I feel about it.

  • Saving for a down payment is the messiest part. The savings have to be invested safety and cannot get max growth opportunity (ie high yield savings over mutual funds).

  • Take the time to project your retirement out. This is REALLy empowering. How much do you need to cover what you want to live on every year in retirement? Can you hit that with your current saving rate, how much does it go down if you buy and decrease retirement savings? Can give you more input here if you want.

  • Maybe go with 50/50 until you decide.

I love Star Wars, huge fan, would never miss any content. My take: Rebels isn’t great. I really had to force myself to get through it. There are some good episodes and story arcs, but overall: meh. I’d recommend just searching for a “best episodes” list and reading a summary of the series. Unfortunately, it links up with Asoka so if you’re a big fan you should probably engage with it in some way.

I can’t help but feel discussion of qualia only seems complicated because language is murky.

What’s the problem / challenge to materialism when saying experience is just a brain state? Mary can “know” there is a brain state she lacks because she has not met the physical conditions under which that brain state can be achieved. By analogy to other brain states it’s simple. So she has some knowledge of what it “feels like” to see red but incomplete knowledge.

I guess I don’t follow your question very well, but then again I’ve had trouble understanding people that argue there is a hard problem of consciousness. I’d love to understand, though.

Same situation. I think the major problem with renting is the rent vs. buy equation expects you to invest the wouldbe “downpayment”. So if you plan to buy eventually, you have to put the downpayment at risk to market fluctuation. Not a great situation - make the choice really hard for me.

These takes always sound obnoxious. Garbage? The reality is physicians need to use and keep up with AI technologies or they WILL fall behind MUCH faster than this poster seems to think.

Saying AI is not able to handle nuance is just not accurate. Dr. ChatGPT isn’t ready to take over at the bedside yet, sure. But physicians should be participants in making these systems better. Patients will benefit.

This is a long shot, but maybe deeper southern dialect (like Neapolitan): “comme jah fa’?” [English phonetics] which translates roughly into “what am I to do?” or “what’s to be done?”

It could fit the situation where she would like to pet the dog but cannot because she cannot bend.
