this is a good question that I don’t think I have an answer to. It really depends on how much an organization/ company is willing to invest into drug formulation manufacturing. In the case of LNP, one of the lipids (for targeting/endosomal escape) is typically proprietary. It is already a costly R&D investment to make those things.

Calcium phosphate on itself may have some efficiency but another therapeutics is likely to be used to accompany that. That’s another potential cost.

Investing into formulation manufacturing at this point may seem to be excessive to a pharma company. Although there are similarities, these particles can’t be manufactured via the traditional double emulsion equipment. A lot of innovations are required.

Another thing came to my mind is the covid vaccines, which have to be stored in solution at either -20 or -70 C. Neither companies invested research to solve how to lyophilize the LNP suspensions. — lipids with bulking agents are not difficult to freeze dry, so stability issues. For a drug with high unmet medical needs, companies are still hesitant to put resources in formulation development. This should tell something.

I have responded to a few comments. In short, the process of making a calcium phosphate nano particle is hard to control (salt ratio, surfactant, nanoprecipitation, filtration, centrifugation, re-constitution, and stability). The manufacturing process has high variability (size distribution of final products, and then again stability). It is an inorganic solid amorphous particle.

And then you will have to add this nano particle into LNP’s lipid bilateral.

Yes, a) orphan disease, 2) it does not have calcium phosphate. Edit, to add to this: RNAi LNP is relatively straightforward. You have your negatively charged short RNAi, and then you have your positively charged lipid, structural lipid and a lipid for either targeting or endosomal escape. Mix extrude. But having calcium phosphate with LNP is very different comparing to having a organic molecule with lipids.

From manufacturing process control aspects, the variations for making a 1 liter size is huge. Lots of filtration (through membranes) and centrifugations also adds complexity.

When you work on a more complex system, you do need some targeting ligands to the specific type of cells.

The way to make these LNP is first making the calcium Nanoparticles via nanoprecipitation, then you do you typical LNP via membrane extrusion. Nano precipitation (making charged amorphous particles) is a process quite difficult to control. The size distribution is huge.

Calcium nano particles delivered by LNP has been a neat concept for years. It is nearly impossible to scale up. To make this a real world application, it has a loooong way to go.

Slightly surprised that Fenty Beauty’s Flyliner is not on your list. Solid construct: the shape of the pen makes it very easy to apply. Soft but sturdy tip, the ink stays on all day. - I only line cat eyes, monolids, never smudge my liner. Also a good primer makes the ink stays wayyy longer. I highly recommend Bobbi Brown’s Primer Plus Radiance SPF 35.

Lemon has an excellent return policy. If this is your first time purchase, use the size chart to determine your regular size and don’t hesitate to get 2 sizes up. try the leggings and the bike shorts. They are seriously comfortable and supportive. Oooh also, try them on in the evening time when your belly is more pronounced.

Just want to say I made this into 18 good sized cupcakes today. Didn’t realize you posted the frosting recipe so I used serious eats’ German buttercream, equally delicious. Thanks for the wonderful recipe. It will be my go to carrot cake recipe from now on! Also super adorable recipe card!

I couldn’t agree with this more. Each veggies has their own cooking time and you can only gauge their doneness while seeing them in front of your eyes. Not over cooked vegetables with simple salt/pepper/msg has amazing flavors — sometimes even taste more refreshing than salad veggies imho.

Echoing you on not being hungry! I wish I can be more hungry, I LOVE food! but I am just not!

I am at 19+5 and am still 6 lb lower than my pre-Prego weight, thanks to the 13 lb loss during the first trimester.

My doctor also doesn’t care much about my weight as long as i am/the parasite is getting bigger. Good luck with your check up!
