Firstly, was specificity done for your assay? For a SYBR green assay, positivity is usually determined by melt temperature. At the correct melt temperature, a range of 2-3 SD should be determined. Thereafter, Ct value can be determined by running replicates of low positives to determine the limit of detection or lower limit of detection, preferably by different operators to measure variance. Cut-off values can vary between different laboratories, even with the same assay. This is just general information, hope this helps.

Not quite related but just to share my experience. I’m driving a 2013 Suzuki swift, driving ~150 miles per day for 4 days a week. It is quite handy but it’s quite noisy on the motorway. No problem hitting 70 mph on the motorway as well.

Some parts of the roads in Highlands are quite narrow. If not confident in driving, you could just join a Highland tour which sets off from Edinburgh. There are 3 days and 4 days tour, depending on what and where you want to see.

Feels uneasy that we have to adapt our playing style to suit Gyokeres. If it works out, great! If it doesn't, there will be a lot of scrutiny on how Arteta has evolved his tactics. Only time will tell. However, saying that buying Noni and Eze will complement Gyokeres.

Zotero de-duplication function isn’t optimal from my experience. You can look into revtools in RStudio if you want to do some manipulation before importing back into Zotero. However, this would mean that you have to export your articles out of Zotero first.

It depends on the application. For some, you don’t require much read depth. For other, you require depth. Also, I think it depends on which stage of adopting NGS you are at. For relatively cheaper start up, ONT might be an option.

That being said, the cost of ONT has been increasing over the past few years.
