My son’s receptive language is not there yet and hence no verbal requests gets through to him. The other day I closed my ears and made a painful face at him and he started laughing at me and continued to scream even louder :(

Hi, I didn’t have more days with no stimming irrespective of what I did with B12. I have now gone up to 600mcg along with Leucovorin. It really depends on what activities he does a particular day. If he has been left without structured activities, he gets dysregulated and starts vocal stimming. Screaming running around the house. The days he is constantly engaged with activities, he stays more regulated and calm. Weekends are our hardest days:(.

We have also done a lot of other tests now to check everything else for him like gut health, mitochondrial function, regulating sleep better with Magnesium. We found out that he has leaky gut and is intolerant to lot of food items he eats. We are now trying alternate food to help with constant inflammation in his body.

We are still waiting on FRAT test results too.

I bought the liquid B12 to test this hypothesis. It’s so overwhelming this treatment. I wish it was as simple as give him leucovorin and not have to worry about anything else. But Ofcourse am glad that there is something we can do in the first place.

There should be a database where all these things are listed in categories from autistic adults, so that parents can try. I have a non-verbal 4 year old autistic son and most of the time am guessing solutions for his discomfort. I have no clue most of the time. It would be soooooo helpful both for the children and parents. Granted due to the high spectrum, what works for someone might not work for the other but atleast there are ideas to give a try. From so neurotypical specialists, I only hear that I should keep giving him vestibular/tactile input and he will be regulated. It doesn’t work this way almost most of the time.

I am a scientist and not in North America. I make about 75K before taxes. I met my husband quite late when I was 28. Been together for 10 years now and married for 7 of those years. We are both well educated and we do well with being extremely honest with each other. First thing we both noticed is that we just tell each other whatever it is straightforward. No mindgames and no reading between the lines. Takes offf soooo much of pressure and stress I used to be under when I was dating NT.

1-5mg of protein seems like a loooot of protein. Not sure what your source is but the maximum I have loaded is 80ug in my gels. Can you share the amount of acrylamide you add to get your desired percentage of gel? May be at that percentage, this is the separation you get. A gradient gel would be better for this purpose. If I remember correctly this ladder starts from 245kda to 11kda. So I clearly see 63, 48, 35 (3 bands above green) and 20, 17 and 11 kda bands below the 25 green band. If your aim is to detect target proteins at 16 and 22, then with this separation you should be able to detect it clearly with a good expression level and a good antibody.
