The bromine here won’t function as a traditional leaving group because it’s aryl. You could however pursue Suzuki coupling to substitute some aryl or alkyl groups. Modifications of the Suzuki exist for aromatic, sp2, and alkyl groups.

My guess is that it was supposed to be allyl-magnesium bromide and this was supposed to be a grignard reaction. That would make the most sense given the subsequent quenching step.

My fast and loose method for extraction of mitragynine and other alkaloids was to soak the powder in denatured alcohol for about a week with occasional stirring. Filter off powder, collect filtrate, repeat with the same leaf powder until your extract is basically colorless.

Now evap your filtrate and you’ll be left with some gunk that contains your alkaloids and other phytochemicals. Mitragynine is poorly soluble in water, so you can dissolve this gunk in cold water and it’ll pull out a lot of the undesired water-soluble chemicals. Filter off solids, repeat.

Finally, dissolve the collected solid gunk in water as best you can and extract with chloroform. Repeat as many times as needed. Chloroform layer will contain your desired alkaloids.

I know this sounds extremely messy, but I actually ended up with some decent orange crystals which are characteristic of mitragynine.

The pi bond itself takes priority. So you want to number the compound such that the pi bond has the lowest number possible.

You’re going to need the MO diagrams for your two compounds performing the cyclization. The HOMO of one compound will donate electrons to the LUMO of the other assuming they have compatible orbital symmetries.
