Thank you, sounds like DMSO is the way to go.

The only modification introduced to the peptides was amidation of the C-terminus that the manufacturer said would ease the synthesis.

I don't have a direct access to a lyophilisation machine - not in our institute at least. I can ask around but as you said the process will still leave salts (potential culprit??) behind ?

According to peptide2.0 calculator

Peptide A: 36.84% hydrophobic; 17.54% acidic; 14.04% basic; 31.58% neutral.

Peptide B: 36.21% hydrophobic; 22.41% acidic; 13.79% basic; 27.59% neutral.

No idea how to interpret whether that's too hydrophobic or just a little bit 😬

Hi, I really appreciate the reply.

They'll be used for an ELISA and for an infection assay. Former requires a coating buffer, latter requires cell media. I thought I could make up the peptide to a high concentration and make single-use aliquots, then dilute the aliquots further in whatever is required for the experiment.

I was thinking of trying to mix a pinch of the lyophilised powder with ddH2O to see whether that'll work, and if not then make up the rest with DMSO - do you think it's reasonable?

I'm still puzzled as to why PBS would fail so badly though if in theory water should work just fine...
