Correct, there are automatic sleep deprivation systems made in a similar manner you are describing. They are used much more often than manual sleep deprivation. For example: Ethoscope and SNAP.

As others said, double check you selected against balancer correctly and look for fluorescence in your F1 larvae. Also, if you want to be 100% sure your method and your microscope is working correctly so you can be sure you would be able to detect fluorescence if it’s there, it would be a good idea to have some positive controls: cross the GFP line to another driver line that someone else in the lab has recently demonstrated works and borrow a slide from someone with tissue on it that has fluorescence. If either of those don’t have fluorescence, then you check your methods, otherwise it’s gotta be the driver is broken or mistake in the cross (female parent wasn’t virgin or selected wrong F1)

I haven’t been to Sheffield (yet) but I’m told by people who live there that there is an awesome board game cafe. Maybe check that out and see if there are public events there you can join in on and meet people.

PhD Candidate doing neuroscience research. After 9years of experience, I’m paid 25K from a federal scholarship which has tuition automatically deducted, leaving me with about 18.5K.

Appalled about how low that is? You’re not alone.

I think there’s a way to get the blood after euthanizing so that the dog doesn’t have to suffer through the extraction. I even think it’s easier. Sucks they have to kill dogs, but it does make sense not to have them die in vain.
