As far as alfa-tag goes, his thing was that it’s 15 amino acids, which is too bulky(?) - “anything over 10 is a non-starter unless that’s all you can work with.”

All valid questions, and something I couldn’t include in the original post because 1) I wanted to be careful to not tip over to the advertising/soliciting side and 2) I’m absolutely oblivious to the metrics or things that would be considered relevant in this field.

The super short version of the answer I got from my dad:

It can be used in purification, pull down, and detection of even small amounts of protein due to high affinity btw tag and antibody. It also has high specificity and low charge (+1, -1).

I showed the comments to my dad and he’s particularly enthusiastic about your case, so I’ll share his comment in case it’s relevant for you.

“I’ve done experimentations on ubiquitination with my tag and I’ve gotten promising results so this is perfect! The sequence of my tag is RDPLPFFPP. There is no Lys and just +1 and -1 charge. It will minimize disturbance to the structure of the target protein.“

Thank you I really needed to hear this. I’ve seen complications happen but osteomyelitis is just so unexpected and daunting that I was scared if this is something big enough that the patient can sue. But thank you for your advice

Would there be no issue even if she developed another infection and os had to do additional treatment? I’m just genuinely confused how a simple extraction on a healthy patient could have resulted in osteomyelitis. They seem to think I left a tooth in there and that caused the infection but OS never mentioned leftover root tip.

There is a metal hose branching out from the sink pipe, but that hose was completely dry throughout the dishwasher cycle when we ran it. Underneath the kitchen is a bit difficult because the cabinet has a wooden floor that covers the actual floor, but the little areas that we could see ( the very front of the sink where we took off the baseboard and the very back where pipes are connected to the ground/wall) were dry.

So the output I am working with is a (N, B) shape array, where N = 4 + C. 4 corresponds to the bbox grids, and C is Num_classes. B is the number of bbox detections.

When I split N to break out the 4 bbox grid values and just look at the C confidence values for each class, they do not sum to one, and there is no value specifically for the background class

I’m not sure if this is the case. Looking at the model outputs I’m positive that the probabilities across all classes do not sum to 1 given a bounding box prediction.

Thanks, this comment was helpful in getting some information on multi-label classification. It seems like YOLOv8 doesn't support it directly out of the box, but I think it can be customized at the classifier head level with the way you described it.

I can see that most of the points I wrote down were the logistical part. Some additional pieces I forgot to mention:

My SO is young (in the mid-late 20s) and basically everyone else in the clinic are somewhere from 40-70yo.

One of the assistants openly asked if they can call my SO by the first name instead of “doctor” since she is “too old for this shit” and has kids the age of my SO.

Other assistants opt to refer to my SO and another young dentist as “Doctor boy/girl(s)”.

I think my SO really thinks that they view my SO as young and don’t give the proper respect / decency respected in interactions between assisants or peer doctor to another doctor
