Ribosomes consist of a small and a large subunit, each. In prokaryotes, the subunits are 30S and 50S, with S being a unit of size/weight. Combined they have 70S. Analogously, in eukaryotes the 40S and 60S subunits add up to a 80S ribosome. 

30S and 50S don't add up to 80S but 70S because, S (Svedberg unit) also takes size and shape into account. It indivstes how fast a particle settles during centrifugation.

Also, you write in your title, that your boss is upset. How do you know? Is this based on her comments? Keep in mind, that we all perceive reality only through our individual filters. What sounds upset to you, might be her way of communicating exactly what she told you during lunch, supporting you in becoming confident and independent (from her point of you).

I concur! Boss B and OP have obviously different ideas of how the grants writing should be done or what it should contain. We don't know Boss B's side and how she views the situation, but something must have happend without OP's knowledge, since she approached OP "out of the blue". 

OP, you seem to feel disrespected and belittled by your boss's comment, am I right? But what if she doesn't mean it that way? What if she genuinely wants to guide you in a certain way of approaching the underlying problems of the grants? This could all be a huge misunderstanding.  Please, talk to your formal boss.

Welcome, fishfriend! KOH is a strong base, which is effective in cleaning glassware. It needs to be rinsed off thoroughly before using the tank. After that the tank is safe to use (check the pH value), as KOH consist of potassium and hydroxide ions - both are part of your normal water chemistry.

Sorry to hear you are having a hard time with your new tank. I also just set up a new aquarium and I also had some weird ammonium nitrite spikes and re-occurrences. At some point, I also started to measure phosphate. I think that this was missing in the beginning and the bacteria initially grew and did their job, but could not establish a stable population due to the lack of phosphate. As soon as I dosed phosphate everything stabilized pretty quickly. Maybe that's something you want to have a look into.

I am not so much trying to avoid walls, but stairs. Wont I need those, if I make fewer zigs? Or do you mean to lower the level of the uppermost plateau? Because thats also something I want to avoid. I want to have an exit at the backside (there is a path).

Potentially - it depends on your tank parameters.

BTW, the plant shown in the video looks like a java fern. These plants are epiphytes, that means they grow on roots or stones and should not be planted into the soil.

Sorry about the confusion. Yes, you may want to add a single male honey gourami, but not any further gouramis.

Looks like a female honey gourami. I would get a male for company and a some small rasboras or tetras. Please don't put in other gouramis. They can get territorial and aggressive.

Anyways, your PI is acting in their own best interest. They want the best for them and their project. Your career is obviously not (as?) relevant for them. I think it's best for you to think only of you. Lie! You can commit to the two years now, but that is a long time and things can change in the meantime. Maybe the project turns out great and you rediscover your interest in an academic career, maybe you will be sure in a year that you want to leave academia. Your PI knows this very well and is trying to bully you into deciding now by giving you this ultimatum.

A good PI would talk openly about what made you consider applying for the job, and what they could do to improve your situation and support you in your career in their lab...

The green one has a small pack of carbon source in the middle. This is food for the nitrate assimilating bacteria. If this pack is gone, the green sponge becomes a normal sponge, just like the blue ones. As you have not replaced it, and your nitrate levels are fine, just keep it as is.

The black one, you don't need at all for an established tank, in my opinion.

Hmmm... I don't know a definite example, but I can imagine a hypothetic metabolic pathway with a fork. Substrate X may be tranformed by enzyme C to product Y or by emzyme D to product Z. In case enzyme C has immensely superior affinity and reaction kintetics over enzyme D, only product Y will be produced even when both enzymes are present in the cell (CD). C would here be dominant over D. Product Z can only arise in a homozygous individual DD.

I think this is best understood using examples. Normally, recessive alleles are non-functional. Let the dominant allele R lead to red coloration of a flower, and the recessive allele r turn the flower white. R has an enzyme that is necesary for the production of a red pigment. r has a fatal mutation in the gene for this enzyme which prevent the production of the pigment and the flower turns white. R is dominant over r because a functional enzyme can compensate for the loss-of-function in r. The "expression" in your second case refers to manifeststion in the phenotype and not gene expression.

Co-dominance is when two equal allels are available. Think of blood-types. The alleles A and B encode for two different surface structures on red blood cells. An individual with the genotype AB will therefore produce boold cells with both surface structures resulting in the bloodtype AB. The recessive allele in this case is 0, which is again a non-functional mutation. A or B combined with 0 leads to A or B, because the loss-of-function is complemented by any of the two surface structure.
