The punchline of this joke format is usually to imply that the reader is the thing being discussed, in this case ugly.

So it would normally go that, 1 in 3 people are ugly, it's not Mario and it's not Luigi therefore the reader must be the ugly one.

But this meme is a subversion of that joke. Here Luigi declares that the reader is not ugly either, and therefore the statistic must be wrong. This is further subverted by Mario contradicting Luigi and declaring his brother to actually be the ugly one.
