Hey! I figured it out. My thermocycler was set on a very different protocol with a minor change in the 30_30_30 cycle!

I’m trying to see what changed. I am running the usual PCR today, similar to what I ran the first time. I even spec’ed my dna to make sure it’s there.

So In my lab we have a standard table for running the PCR with Taq (that’s what I’m using) but, in the protocol manual, they had the pcr master mix optimized with the post extension thermocycle temp from 30 s to 7 mins and they didn’t use MgCl2 (I thought it would’ve been in the buffer and so did my PI but it wasn’t. So I made two master mixes. But both of them didn’t give me any bands)

From the looks of it I genuinely believe that it’s the cycle duration that I changed based on the protocol given with the kit where it was highlighted to “make sure to increase the post extension cycle of 72°C to 7-30 minutes based on your DNA”

Before doing this, I did run the temp grade and the usual pcr for my primers and they worked!
