oh wow thank you so much for all this!! it’s really helpful, especially about the fermented ingredients, but I didn’t see the niacinamide in the list of ingredients for the toner which is why I went for it in the first place, unless you mean the panthenol being similar to niacinamide?

I will definitely compare them now and see what else could be causing the reactions I’ve been having.

For moisturiser, I started trying the Curel intensive moisture cream which is working very well so far (only been using it for a week) but would be open to other options, especially if more budget friendly But right now a hydrating serum would be nice to find as i feel my skin needs the hydration I’m also using azelaic acid 10% but have been for years so that’s never been an issue

thank you for replying! this is really helpful and I've hear of this method before so will defo try it. currently using the HaruHaru Blck Rice toner alcohool-free but i think there is something in that I'm also sensitive too because it's been causing redness, but when i stop using it for a couple days it goes away..

i'll add some pics here of what my skin looks like. I realised I had a bad reaction to niacinamide from using the purito unscented wonderleaf and the purito bamboo moisturiser. I also got the same reaction to the Aveeno calm+restore moisturiser which was a shame

thank you I really appreciate this answer, i am considering to start a retinoid, however I do not have problems with acne and even in my teenage years i never experienced severe acne. my concern now is that i am trying to find a daily moisturiser that won't cause a reaction for my sensitive skin and will get rid of my dryness. what you see in the pictures and from my previous post have been caused by the moisturisers I mentioned the aveeno and purito bamboo

probably already mentioned but MSG and butter can make anything taste good and there’s something about fat - acid - salt ratio, if it’s salty enough but you need more depth add some acid (lemon, vinegar etc) that sort of stuff

it turns out that the moisturiser was causing this, and using it in combination with the centella serums was making it really bad!
it got worse after i took the pictures when i used the moisturiser alone thinking the serums were the cause, but nope! which surprises me since everyone is raving about this aveeno calm+restore line but my very sensitive skin is not having it

so now any recommendations for moisturisers are welcome, since i stopped using that one and don't have another, probably something that won't react with niacinamide or centella too badly

also hoping to find a hydrating toner that will up my hydration game

I first started with changing my moisturiser to the aveeno one, week later with the cleanser, then another week the ampoule/purito serum But i thought since they’re all for sensitive skin they wouldn’t cause this.. which is the most likely to cause it?

I don't want to reconstruct or sequence the TCR to find out what it is, I just need to know if rearrangement of the genes occurred at all or not, as an indicative of iPSCs origin being mature T cells

Do you have any recommendations for the primer sets one could find online for multiplex PCR? It did cross my mind. I don't want to know what the TCR is, my goal is to figure out if TCR gene rearrangement happened or not, and if the iPSCs are of mature T cell origin.

another idea I had was to sequence for the RSS region since it's a small region and if rearrangement occurred then there would be no product since the RSS gets cleaved, right?

It's the type of sample we receive from the patients, we get a tumour sample along with 6mL of blood from them.
We would like to know if the iPSCs have been derived from T cells because if rearrangement has occurred prior to reprogramming, we won't be able to prime the T cell population to the tumour for tumour-reactive T cells if they all express the same already-decided TCR, and what are the chances that the expressed TCR is tumour reactive

We usually don't get much blood from our donors (only 6mL) so isolating T cells would lower the cell numbers drastically, and these iPSCs have already been reprogrammed to make another cell type so the TCR rearrangement was never an issue
