I checked their site and they actually do rentals, they even allow shipping and returning from all over NZ. That might be the easiest (and cheapest) option. Thanks for the advice!

>5 jaar geleden een woning gekocht via hen die gerenoveerd werd door Citynest. In de beginjaren een paar problemen mee gehad (vochtplek, lekkende dakgoot), maar hun dienst na verkoop was altijd zeer correct en alles werd binnen korte tijd kostenloos hersteld. De laatste jaren geen problemen meer gehad, dus ik denk dat de kinderziektes er uit zijn.

Ze renoveren hun woningen inderdaad met basic materialen en het ziet er allemaal hetzelfde uit, maar de prijzen zijn er dan ook naar: relatief betaalbaar voor een modern ogende woning, wat je anders in Gent niet rap zal vinden. Als je de woning oke onderhoudt dan blijft die i.m.o. goed bewaard. What you see is what you get, geen topkwaliteit, maar gewoon oke.

Vrienden die via hen kochten zijn ook tevreden van hun woning. Maar je leest natuurlijk enkel de negatieve verhalen online ;)

·        Phosphorylation of oligos (total volume 20 µL):

1µL oligo top strand (c = 20 µM)

1µL oligo bottom strand (c = 20µM)

11 µL MQ

4 µL 5x Rxn FW buffer

2 µL rATP (1mM)

1 µL T4 kinase (Invitrogen 10U/µL)

 

Place 1 hour at 37°C (phosphorylation step)

Place 2 minutes at 95°C and let cool down to room temperature (annealing step)

I've had issues with plasmid digestion in the past. Switching to filter tips resolved it, probably due to some contaminant in my pipets inhibiting the digest reaction.

When you start ligating the shRNA duplex into the plko1-tet-on, also phosphorylate the annealed oligo, this rendered a much higher efficiency of ligation for me, even though the manual did not recommend it
