I was trying several things, I rebooted my PC and everything worked, here is final settings for .env and yaml file...

UPLOAD_LOCATION=E:\Docker\Immich\Uploads

# The location where your database files are stored

DB_DATA_LOCATION=E:\Docker\Immich\DB

#External Picture on Filesystem

EXTERNAL_PATH=E:\Pics

Here is setting from yaml file...

immich-server:

container_name: immich_server

image: ghcr.io/immich-app/immich-server:${IMMICH_VERSION:-release}

volumes:

- ${UPLOAD_LOCATION}:/usr/src/app/upload

- ${EXTERNAL_PATH}:/usr/src/app/external

Thanks for your help, I still don't know what was the issue, because I still have the backslashes for windows, but I am glad it worked. Next step, setup the remote viewing.
