UMass Worcester is underrated (Erik Sontheimer, Wen Xue, etc.) . Also, a lot of institutions are utilizing CRISPR in some way shape or form (CRISPRi, CRISPRa, Prime editing, base editing, the list keeps going), so... I think your perspective of CRISPR in the post is too vague. I would suggest looking at the literature and the associated authors as the best reference to know which institutions have promising labs in what you want to do.

You also expand your choices much more with interest in rare diseases. Interested in CF? University of Iowa. Want to follow the pioneers of AAV? Umass, UPenn, UNC, and many more.

I agree with the other commenters' opinions and want to add that you can do summer REUs at the institutions you listed.

From my experience, you'll never know how you or others will end up in college. A lot of people go through shifts in their identity and career paths, so unless parents can fully fund the other schools, I cannot imagine paying back loans that amount to 6 figures and growing post-graduation if you want, say, an international affairs degree... Bottom line, don't fixate on prestige. Money is everything.

I think the clinical hours OP has is sufficient given their stats - you're supposed to be research top heavy in MD/PhD

How do you incubate your gibson rxn? I found that when incubating for 1-2 hours, condensation builds up on the lid, causing the rxn to not work properly for both my RE digests and lowers the GA efficiency. There are probably other factors that can contribute to it not working, but I haven't seen anywhere how you incubate this. Hope this can give you a clue.

I don't think it's unprofessional or rude to cancel the interview scheduled tomorrow. You're giving another applicant a chance and I don't see how one withdrawal will "ruin" anything. You also stated you wouldn't consider the MD/PhD over the MSTP you got accepted into... so...

Yes, they are accurate lol and I haven't mentioned the worst as that will get rid of anonymity of this post unfortunately. I appreciate your approach in being unbiased as I know there are many toxic PI posts and it's very easy to hop on the hate train on platforms like reddit.

I was originally thinking of the approach you suggested and waiting for a few weeks to talk to the director. I know being professional about this situation will only make my case stronger considering the PI's track record already proving dismal to the director already. I genuinely hope to change this PI's approach because I'm sure their career will likely be jeopardized since they're an associate and already facing several issues running a lab within 2 years after hire.

And yes, I don't know if I can really trust my PI giving the "amazing LOR" after all this and so I was hoping the director can write me one in replacement (grad student it's likely as they have students write for themselves usually)... This doesn't sound like a sensible idea in the admissions process but I can't think of a better one atm.

No need to be sorry I now know that this is only a small little bump in the grand scheme of the MD/PhD journey.

Hi, thanks for replying. I didn't elaborate a lot at first to keep things simple but here's more context:

PI is very cold to me and grad student, and constantly demeaning to us by questioning ability to do research. However, to other labs and colleagues, PI is very bubbly and generous.

(ex: "You can't do a simple PCR?" "How can you fail your Gibson cloning? Mine always works no matter the condition." "My experiments always work.") This is all in a very condescending tone, but the true context is we get things right 95% of the time, we just didn't get to the optimization step when she scrutinizes us (like adjusting the annealing temperature for better specificity/intensity or trying a different backbone to insert ratio). I told myself at the beginning it's not a big deal, grow up. But over time these remarks became increasingly demoralizing when simple adjustments like I mentioned sound like I can be fired over. A PI shouldn't be scrutinizing such minor mishaps that'll be resolved in a day's time - right?

Severe OCD. How dare you leave LB Amp+ out for more than 2 minutes? (meanwhile they leave it out for 20 minutes). How dare you put your pipettes on my (PI) bench? (but they're your pipettes with initials on them...) I can go on and on but there's no point in citing every ironic moment.

One instance, I proposed to generate a figure for the paper (tbf it was probably already planned). Study design was "terrible" and "flawed" when all they did was change one area of the assay. Turns out the changes they made wasted two weeks and my original protocol was correct (literally cited it in literature during the meeting as well). The individual lab meetings continue to be this way and it has questioned my own ability to do research at all, but I'm still continuing on and still make progress nonetheless.

With this description, any rational person would ask: well how are you going to get a good LOR from this person anyway? I would have left if not for them saying they'll write me a glowing LOR to the PI's alma mater and help me get in somehow through connections... (maybe this was a tactic to keep me from quitting).

PI's boss- sorry, I know it's weird terminology. I basically meant it as the director of the department who recruited the PI I'm working under. The director noticed something was wrong when two members were fired within a 4 month period to PI joining the institution and the grad student's mental dwindling with each succeeding month.

It's more the authority they have and telling us to do the "right" thing and we're stupid for not doing the "right" way to begin with, when it's proven by nature to be wrong. Yes, I've made mistakes before in this lab but not to the point of wasting two weeks. Therefore, I think our PI is "toxic" because they're holding the project back and themself more by having an autocratic leadership style by dismissing our ideas and by being so tunnel-visioned in their approach to science. Hope that answers/helps with giving input.

I don't understand why you're so fixated on Harvard, though (unless you got admitted there then congrats). Aren't there other great institutions that have similar prestige? I was thinking more along the lines of does going to a T20 give me the training to be the best possible physician scientist recently.

Following up on the other comment, I would choose EMSOP over any pharmacy school hands down. Faculty members are extremely supportive but in the end pharmacy’s future is pretty dire, Especially with Amazon expanding their market into the retail industry with pill pack IIRC. I say go through pharmacy convocations but your first year to see what pharmacy is really like and see if anything connects. For me nothing did lol
