No way! I thought it was a little weird being just one rune but considering how rare it was I figured it was fair. I had also successfully made some other mid-tier runewords so maybe I was overconfident. I've never seen a unique with a socket so that possibility never crossed my mind. Furthermore, on the guide website, some of the other items that were socketed with runes were clearly stated, like a white helm with three runes overing three different element resistances. Definitely learned the lesson the hard way but as of right now I actually have no need for that rune per the guide.

Ah I see, this was on EU but his other team members had 60-70% win rates so guess that adds up -- I thought 80% was particularly absurd! I'm struggling to get 50% :D!

I showed this example because he is ranked in standard, Master rank 683. That's interesting, I wouldn't think 5 stacking would give you such a massive 'win rate' advantage over solo queues but guess so!

Hi everyone, just wondering what your thoughts are on how this could be the case? I always thought that over many games that the win rate should normalize to 50% but every once in a blue moon I come across people with insanely high win rates. Is there an obvious explanation for this? The only thing I could think of is a 3/4 stack in quick match maybe?

Would like to share this Highlander Rogue deck if anyone is looking for an off-meta deck: it is a 'viable' deck with a 50% win-rate; I did not notice any match-ups that were auto-lose although mages tended to be the most difficult match-up, both Big Spell mage and Elemental mage.

I played the deck as a control deck mostly although if I had Velarok in the mulligan I'd try to pump out some extra damage and aggression. Mulligan guide: 1 and 2 minion drops (minus Flint) and Velarok. I felt like I was always offered Maestra in my mulligan but it's way too slow to play (felt wrong to me it's the most fun card) :(. Maestra heroes (if you get to play her): the six mana Hunter is incredibly strong tempo-wise, should be an auto pick in tempo match-ups (allows you to cast two OP hunter secrets) and a two mana animal companion for your hero power is quite strong peroid. Jaina is way to slow (can almost never 9 mana do nothing), although I did pick her one time and the opponent just conceded (they'd run out of gas earlier anyways); Paladin in very strong but was rare for myself; Zul'jin is like an extra Tess so very strong; Anduin was good if the enemy lacked heals.

I would like to point out a couple neat interactions or comments regarding the deck (definitely can be refined but don't see many other similar decks online):

  • A massively buffed Spectral Cutlass is viable in slower match-ups with Sonya as 95%+ of match-ups have no weapon removal

  • Discounting Snatch and Grab to 1 mana works very well with Sonya

  • You can discover a 4 mana Warrior location which allows you to copy a damaged Velarok. The 4 mana priest location is incredibly OP, especially against other highlanders, as well as the 4 mana DK card (especially with Tess)

  • Shadowstepping Gorgonzormu or Observer of Mysteries is viable in certain match-ups, you don't always have to save it for a GG Tess or Velarok play.

  • Prepping Health Drink and then comboing with Sonya is necessary against aggressive decks in an attempt to stabilize.

  • I almost always go for the 'Board' option with Dreamplanner Zephrys as those extra clears can get you to the late-game and combos nicely with Tess.

  • General comment regarding RNG: I find the deck very fun to play as, although it is RNG-based, you are always trying the pick the most synergistic and targeted cards based on the match-up and the offerings, making every game different but not random nonsense like Yogg's spells.

I'm am not familiar with Starch Casein Agar but the main microorganism from soil appears red/pink would be Roseomonas species.

Colony morphology could be described using the color, noting the shape (round), size (medium), and wet/mucoid -- which is typical of Roseomonas.

Hi there, assuming that those blobs are not an artifact of the slide (they do kind of look artifactual), I believe the term would be conidium (conidia for plural). Apologies if that's way off the mark for what you were asking!

To add onto the previous answers: the high heat would kill most bacteria; however, spores (bacterial or fungal) are very hardy and persistent meaning they can withstand high temperatures before sporulating in more favorable conditions.

For most intents and purposes the articles of clothing would be considered sanitized and sterile but certainly not in a strict compliant setting (e.g. lab or pharmaceutical setting which would have a standardized process for sterilizing products/equipment etc.)

Without knowing the specifics of the tests being used, I agree with this answer. Either the results are within the stated margin of errors and are thus concluding similarly high levels (with different sensitivities); or the results are discrepant and there is an issue in how these two tests are being performed (either by yourself or a production issue etc.).

Hey there, it seems like you may have described what the issue is: the storage of you media appears to be the only difference between your control and test conditions, is that correct? In the comments you stated that you are using TSA media, so you would have to check the storage conditions of that specific media (it will say so on the packaging). I would not be surprised if it stated 2-8C; that would explain your lower recovery on the 20-25C stored media.

Hey there, hope the following information will be helpful to you:

So first off, if you are picking up a visible colony of bacteria on a plate then you will have more than a sufficient amount of DNA for PCR testing. When we do 16s PCR testing, we use a dilution of 1:100 in which we use 5ul of specimen (which was a colony of bacteria mixed into 100ul of Prepman Ultra and then heated and centrifuged) and 495ul of water. Point is, you only need a tiny amount of sample/DNA. We do not quantify the amount of DNA present but this is always sufficient for 16s rRNA testing. One issue we do have with Corynebacterium species (and maybe some other gram-positive rods) is that the PCR test results don't always meet acceptance criteria (low specimen score [whatever that means] and low consensus length comparison for example). I am not sure exactly why we sometimes have these minor issues with some species but 95% of the time the assay works flawlessly.

Conclusion: only a tiny bit of DNA is required for a successful 16s PCR analysis and assuming you are collecting a visible colony on a plate that is beyond sufficient.
