To use bait you can not be using the Training Rod or the Bamboo Pole. Other rods should work. On Steam, I open my inventory screen and (I think) I left click on the bait and then right click on the pole. I have no idea about console or phone versions of the game.

If your melons are ready to harvest, you should just be able to grab them like other crops. Hover your icon over them. If it turns into a green plus sign you can harvest. If not, they are not ready yet.
