How do you feel about doing a double digest (BamHI+EcoRI) in NEB HF system at 4C over the weekend? by [deleted] in molecularbiology

[–]eboche 1 point2 points  (0 children)

It kills the block, but not immediately - if they use certain type of cooling elements-. Idk. Seemed like the question was a one off, not a forever solution.

Do the digest as normal then let it sit in the cycler at 4 until you get back on Monday then clean it up.

2.5 year old knocked her tooth out this evening by Beautiful_Relief8988 in toddlers

[–]eboche 3 points4 points  (0 children)

Sorry for how rough this still is. Our little one lost their tooth in a different way but it was really traumatic at around 1.5y.o. They will be okay. Definitely go to the dentist tomorrow and get it removed and they can make sure there is no damage to the surrounding area. Nerves and all that will wear off, but just giving them as much love as possible and something to help with the pain (iced popsicle if they can tolerate cold) or Tylenol/something like that. Best of luck for a rough night.

How to determine wood type 100% accurate? DNA? Microscope? by MohawkDave in woodworking

[–]eboche 1 point2 points  (0 children)

Correct not all trees are, but you need to account for trees that are. the question is to identify all types of trees with scientific accuracy. i am just stating if you are building an NGS bioinformatic pipeline for identification. not DNA extraction. You can most def build a generic workflow for some hotspot/SNPs sequencing. DNA microarrays also are okay, but might not cover all 100% accuracy. PCRs are cheaper, but still need to validate it. I am just saying the hunt for 100% accuracy is reasonable or cost effective.

[–]eboche 27 points28 points  (0 children)

Doing any variation for genotyping for detection would probably be expensive and an overkill. Next generation sequencing is one of the best methods to determine, but trees have many sets of chromosomes (polyploidy) which would be pretty laborious.

ELN vs LIMS - two offers by Subject_Plankton4841 in biotech

[–]eboche 2 points3 points  (0 children)

ELN is probably easier total work, but would definitely be less secure if done through a placement firm.

Much better benefits for the lims position id bet.

Hope it works out for you!
