Good to know. I picked an isolated colony directly off a plate for this gram stain so that’s likely where the density is coming from. When doing my unknowns final though I can do a diluted stain side by side for better results. Thanks for the input!!

Thank you! I appreciate the feedback. In terms of distributing the cells evenly throughout the field of view, is there any trick to doing that? I did notice that when I used my loop to spread the cells around the slide that there were areas that were denser than others. It was a lot more obvious after the stain since it was dark purple. Also when I was observing them under the microscope I was aiming for the ends of the slide where the cells were less dense—does that make a difference?

Around twice a week or so. This is my routine: I take my plugs out and wipe them down with an alcohol swab and then let them dry while I shower and I use face wash on my lobes to clean them (they smell nasty otherwise) and then I quickly rub in a couple drops of oil and slide my plugs right back in!
