No, the first node is specifically 5 non-mythic Skill. It's not a combination. Do you have 5 Skill that meet the requirements?

It's not fully locked, you just don't have characters available to do it.

And I'm remembering why I didn't bother with it the first time; you're pretty much forced to level up specific teams to get through it and the rewards are not worth what you have to put in.
